不同产地及炮制方法雪上一枝蒿多糖含量分析

摘　　要：目的对雪上一枝蒿中多糖的含量测定方法进行研究,建立紫外分光光度法测定雪上一枝蒿多糖的含量测定方法,并考察不同产地、不同炮制方式对多糖含量的影响。方法用80%乙醇回流去除还原性糖等杂质,后热水提取多糖,采用苯酚-硫酸比色法于488 nm波长处用紫外-分光光度法测得吸光度值,从而得出其多糖的含量。结果雪上一枝蒿多糖以无水葡萄糖计,在0.003~0.015 mg/mL范围内呈良好的线性关系(R=0.9999),回收率为99.35%~99.85%(n=6),RSD值为0.18%。结论本法准确、可靠,重现性好,适用于测定雪上一枝蒿中多糖含量;不同产地不同批次药材中的多糖含量存在一定差异;炮制方式对雪上一枝蒿多糖含量无显著影响。Objective:To observe the expression of HSP60 and Caspase-3 of acupuncture in rats with cerebral infarction and to preliminarily explore the protective mechanism of acupuncture on neuronal apoptosis in rats with ischemic stroke.Methods:The cerebral ischemia rats were induced by middle cerebral artery suture to make reperfusion injury models and randomly divided into sham operation group,model group,electroacupuncture point group,electroacupuncture non-meridian non-acupoint group and moxibustion acupoint group.After the intervention of electroacupuncture or moxibustion,the neurological deficit score and postural reflex score of the rats were evaluated at 0 and 12 hours after the operation.The expression levels of HSP60 and Caspase-3 mRNA were detected by fluorescence quantitative PCR.Results:The nerve function of the rats in the electro-acupuncture acupoint group and moxibustion acupoint group was better than that of the model group,and the difference was statistically significant(P<0.05).Compared with that of the model group,the HSP60 mRNA of the electro-acupuncture acupoint group increased significantly,and the difference was significant(P<0.05)while the expression of Caspase-3 mRNA did not change significantly.Conclusion:Electroacupuncture can improve the neurological function of rats with cerebral ischemia-reperfusion injury.The mechanism may be related to the up-regulation of the expression of HSP60 in brain tissue and mediating anti-neuronal cell apoptosis.
