摘  要:目的:比较β-环糊精(β-CD)、羟丙基-β-环糊精(HP-β-CD)对川芎挥发油(川芎油)的包合行为差异性。方法:制备川芎油β-CD、HP-β-CD包合物,以川芎油中主要成分洋川芎内酯A和藁本内酯为模型药物,相溶解度法考察包合物的平衡常数K和增溶倍数,并进一步考察其热稳定性。结果:在0~15 mmol/L范围内,川芎油中洋川芎内酯A和藁本内酯的溶解度随着HP-β-CD浓度的增加呈线性增加,包合类型为AL型,随着β-CD浓度增加呈非线性增加,包合类型为AN型。60℃条件下,两种包合物均能提高川芎油的稳定性,β-CD包合物中洋川芎内酯A和藁本内酯10天稳定性的相对保留率分别为100%,98.66%,HP-β-CD包合物相对保留率分别为96.59%,90.36%。结论:川芎油与β-CD、HP-β-CD的包合行为存在差异,HP-β-CD能明显增加川芎油溶解度,β-CD包合物的稳定性优于HP-β-CD包合物。Objective:To compare the inclusion behavior difference ofβ-cyclodextrin(β-CD)and hydroxypropyl-β-cyclodextrin(HP-β-CD)to Chuanxiong volatile oil(Chuanxiong oil).Method:Theβ-CD and HP-β-CD inclusion complexes of Chuanxiong oil were prepared.The main components of Chuanxiong oil,Ligustride A and Ligustilide,were regarded as model drugs,to investigate the equilibrium constant K and solubilization multiple of the inclusion complex with phase solubility method,and further investigate the thermal stability.Result:In the range of 0~15 mmol/L,the solubility of Ligustride A and Ligustilide in Chuanxiong oil increased linearly with the increase of HP-β-CD concentration showing the inclusion type of AL,and non-linearly increased with the increase ofβ-CD concentration showing the inclusion type of AL.AN 60℃,both inclusion complex could improve the stability of Chuanxiong oil.The relative retention rates of 10-day stability of Ligustride A and Ligustilide inβ-CD inclusion complex were 100%and 98.66%.The relative retention rates of HP-β-CD inclusion complex were 96.59%and 90.36%.Conclusion:There is difference in the inclusion behavior of Chuanxiong oil withβ-CD and HP-β-CD.HP-β-CD could significantly increase the solubility of Chuanxiong oil.The stability ofβ-CD inclusion complex was better than that of HP-β-CD inclusion complex.
