摘  要:目的:观察复方新会陈皮含片(Compound Xinhui Chenpi Lozenge,CXCL)的止咳祛痰作用及初步机制。方法:采用网络药理学方法,结合通路富集分析,预测含片与呼吸道疾病可能的通路及靶点;采用氨水引咳、酚红排泌、气管排痰实验和气管纤毛运动等多个实验,观察复方新会陈皮含片的止咳、祛痰作用;复制大鼠急性支气管炎模型,观察大鼠造模及给药前后的肺组织病理结构、水通道蛋白AQP4、APQ5及粘蛋白MUC5B的mRNA变化,屏障蛋白MLCK、ZO-1及凋亡相关蛋白Bc12、Fas的表达变化。结果:网络药理学富集结果显示复方新会陈皮含片的作用主要涉及炎症、钙离子信号、肌动蛋白调节、凋亡自噬信号等。复方新会陈皮含片给药后咳嗽潜伏期小鼠数量明显减少,且均能显著降低5 min内的咳嗽次数。气管酚红的浓度在给药后显著升高;增加大鼠毛细管排痰量;提高离体兔气管纤毛的运行速率。在急性支气管炎大鼠中,模型组大鼠肺部支气管上皮细胞肿胀,管腔内分泌物增多,给药后恢复;支气管内粘蛋白分泌增多,而给药后减轻;水通道蛋白AQP4、AQP5及粘蛋白MUC5B mRNA相对表达升高,给药后降低;肺部屏障相关蛋白MLCK表达增加、ZO-1表达降低,给药后恢复;抗调亡蛋白Bcl 2表达降低,凋亡调节分子Fas表达升高,而给药后Bc12表达升高,Fas表达降低。结论:复方新会陈皮含片具有止咳化痰的作用,其作用机制可能调节肌动蛋白、抗凋亡等改善屏障功能有关。Objective:To observe the antitussive and expectorant effects of Compound Xinhui Chenpi Lozenge(CXCL)and preliminarily investigate its mechanism.Method:The network pharmacology method combined with pathway enrichment analysis was used to predict the possible pathways and targets of CXCL on respiratory diseases.The effects of CXCL on relieving cough and eliminating phlegm were observed in experiments of cough induced by ammonia,phenol red secretion,tracheal expectoration and tracheal cilia movement.The rat model of acute bronchitis was established and the pathological structure of the lung tissue,mRNA changes of aquaporin AQP4,APQ5 and mucin MUC5 B,as well as the expression changes of barrier proteins MLCK and ZO-1 and apoptosis-related proteins Bcl2 and Fas were observed before and after modelling and drug administration.Result:Network pharmacology results showed that the effects of CXCL mainly involved inflammation,calcium signal,actin regulation,apoptosis autophagy signal and so on.After CXCL administration,the number of mice in cough latency significantly decreased,and the number of coughs within 5 minutes decreased significantly.The concentration of phenol red in trachea increased significantly after administration.The capillary sputum excretion of rats was increased.The running speed of rabbit tracheal cilia in vitro was improved.In acute bronchitis rats,the bronchial epithelial cells in the model group were swollen with increased secretion in the lumen,which was recovered after administration.The increased endobronchial mucin secretion was decreased after CXCL administration.The relative expression of aquaporins AQP4,AQP5 and mucin MUC5 B mRNA were increased in model and decreased after administration.The expression of lung barrier-related protein MLCK was increased and ZO-1 was decreased,which were both recovered after administration.The expression of anti-apoptotic protein bcl-2 was decreased,and the expression of apoptosis regulator Fas was increased in model,while the expression of bcl-2 was increased an
