摘 要:从青岛市市南区栈桥海滩采集的底泥和水生植物中获得微生物进行驯化研究,以期筛选出高效降解纤维素的耐盐型菌株并应用于含纤维素高盐度废水的处理中。使用盐度为3%的含纤维素模拟废水对微生物进行梯度驯化,CODCr去除率最高达62.4%。运用选择性培养基和刚果红染色法对菌株进行初步筛选、DNS法测定FPA酶活进行复筛后,得到4株具有良好处理效果的耐盐型产纤维素菌株。4株菌株在30℃,pH=7的外界环境条件下生长良好,其FPA酶活分别为0.047,0.052,0.065,0.120 U·mL^(-1)。In this dissertation,microorganisms were obtained from substrate sludge and aquatic plants collected from Zhanqiao Beach,Shinan District,Qingdao City for domestication studies,with a view to screening out salt-tolerant strains that efficiently degrade cellulose and apply them to the treatment of cellulose-containing high-salinity wastewater.They were acclimated using a cellulose-containing simulated wastewater gradient with a salinity of 3%,and the CODCr removal rate was as high as 62.4%.After preliminary screening of the strains with selective medium and Congo red staining method,and rescreening of FPA enzyme activity by DNS method,4 strains of salt-tolerant cellulose-producing strains with good treatment effect were obtained.The four strains grew well at 30℃,pH=7,and their FPA enzyme activities were 0.047,0.052,0.065 and 0.120 U·mL^(-1).
