脂氧素A4对横纹肌溶解大鼠横纹肌的保护作用

摘　　要：目的:研究脂氧素A4对横纹肌溶解大鼠横纹肌的保护作用及机制。方法:SD大鼠45只,随机分为3组,每组15只,包括正常对照组、模型组(肌注50%甘油10 mL/kg)、脂氧素A4组(模型组基础上加1μg/kg脂氧素A4腹腔注射)。72 h后取大鼠下肢肌肉标本进行病理检查,Western Blot法检测肌组织核转录因子(NF)-κB的蛋白表达。同时检测血清白细胞介素(IL)-6、肿瘤坏死因子(TNF)-α水平。结果:模型组TNF-α、IL-6水平分别为(214.03±15.52) pg/mL和(131.92±6.31) pg/mL,而正常对照组分别为(61.36±4.96) pg/mL和(48.52±4.00)pg/mL,差异均有统计学意义(P<0.05),表明模型制备成功;和脂氧素A4组[分别为(93.78±5.25)pg/mL、(67.28±6.23) pg/mL]相比较,差异也有统计学意义(P<0.05)。与模型组比较,脂氧素A4组肌组织NF-κB表达明显下降(P<0.05),病理切片中横纹肌组织损伤显著减少。结论:脂氧素A4可减少横纹肌溶解大鼠血浆IL-6和TNF-α的释放,抑制肌组织NF-κB蛋白的表达,从而对横纹肌溶解的大鼠的横纹肌起到保护作用。Objective: To study the protective effect and mechanism of lipoxin A4 on striated muscle of rats with rhabdomyolysis.Methods: A total of 45 SD rats were randomly divided into 3 groups(15 rats in each group): normal control group, model group with intramuscular injection of 10 mL/kg glycerol,and lipoxin A4 group(intramuscular injection of 10 mL/kg glycerol + 1 μg/kg lipoxin A4 intraperitoneal injection). After 72 hours, the lower limb muscle specimens were taken for pathological examination, and the protein expression of nuclear factor(NF)-κB in muscle tissue was detected by Western Blot. At the same time, the plasma interleukin(IL)-6 and TNF-α levels were detected in all rats.Results: The levels of tumor necrosis factor(TNF)-α and IL-6 in model group were(214. 03±15. 52) pg/mL and(131. 92±6. 31) pg/mL, respectively, which were significantly different from those in normal control group([61. 36±4. 96] pg/mL and [48. 52±4. 00] pg/mL, respectively, both P<0. 05);and compared with that respectively in lipoxin A4 group([93. 78±5. 25] pg/mL and [67. 28±6. 23] pg/mL), the difference were significant too(both P<0. 05). Compared with model group, the expression of NF-κB in the muscle tissue of lipoxin A4 group was significantly decreased(P<0. 05), and the damage of the striated muscle tissue in the pathological section was significantly reduced.Conclusion:Lipoxin A4 can reduce the release of plasma IL-6 and TNF-α, and inhibit the expression of NF-κB protein in the muscle tissue of rats with rhabdomyolysis, thereby protecting the striated muscle of rats with rhabdomyolysis.
