摘  要:本研究通过分析tae-miR167d及其靶基因ARF12在小麦品种济麦22和西农889相同时期不同器官和不同时期相同器官中的表达水平,以及冷胁迫前后8个小麦品种叶片和幼穗中表达水平,以明确tae-miR167d及其靶基因ARF12在小麦中的表达时空分布情况以及不同品种中冷胁迫前后的变化趋势。结果表明,在济麦22和西农889相同发育时期的叶片、叶鞘、茎和幼穗中tae-miR167d和ARF12的表达水平不同,并且随着发育时期的变化,tae-miR167d和ARF12的表达水平也发生变化。分析8个品种正常条件和冷胁迫下叶片和幼穗中tae-miR167d和ARF12的表达情况,发现除临麦4号的幼穗及周麦22和濮麦053的叶片以外,其余品种的叶片和幼穗中tae-miR167d和ARF12表达呈负调控关系;以济麦22的叶片和幼穗为参照,发现多数情况下,叶片中tae-miR167d的表达量低于ARF12,而幼穗中tae-miR167d的表达量高于ARF12。上述结果表明,在小麦的发育过程中,tae-miR167d和ARF12表达水平在不同器官和不同发育时期处于动态变化中,受到冷胁迫后二者呈负调控关系。In this study,we analyzed the expression levels of tae-miR167 d and its target gene ARF12 in the same organs at different development stages and different organs at the same development stage of two wheat varieties Jimai 22 and Xinong 889.The expression levels of tae-miR167 d and ARF12 in leaves and young spikes of 8 wheat varieties were also analyzed before and after cold stress,so as to clarify the temporal and spatial distribution and the changing trends of tae-miR167 d and ARF12 in different wheat varieties before and after cold stress.The results showed that the expression levels of tae-miR167 d and ARF12 were different in leaves,leaf sheaths,stems and young spikes of Jimai 22 and Xinong 889 at the same development stage,and also changed in the same organ with the promotion of developing stages.After cold stress treatment at the stage of female and stamen differentiation,the expression levels of tae-miR167 d and ARF12 were negatively regulated in leaves and young spikes of the varieties except for young spikes of Linmai 4 and leaves of Zhoumai 22 and Pumai 053.With the leaves and young spikes of Jimai 22 as control,it was found that the expression level of tae-miR167 d was lower than that of ARF12 in leaves but higher in young spikes under most circumstances.The above results indicated that the expression levels of tae-miR167 d and ARF12 were in a dynamic changing process in different organs and different developing stages during the development of wheat,and their expression showed negatively regulated relationship after cold stress.
