摘 要:目的:观察神经根磁刺激对慢性不完全性脊髓损伤大鼠髓鞘修复的影响。方法:将18只雄性SD大鼠按随机数字表法分为假手术组、模型组、治疗组,每组6只,模型组建立钳夹型脊髓损伤大鼠模型,治疗组于脊髓损伤术后第11天开始为期14天的治疗。治疗结束后处死大鼠,采用实时荧光定量PCR检测损伤处脊髓组织脑源性神经营养因子(BDNF)、神经营养素-3(NT-3)、胶质纤维源性酸性蛋白(GFAP)的mRNA表达;采用Western Blot方法检测髓鞘碱性蛋白(MBP)的表达;采用免疫荧光染色观察损伤区MBP的表达;采用LFB髓鞘染色(Luxol Fast Blue,LFB)观察损伤区髓鞘的病理变化。结果:神经根磁刺激治疗后大鼠BDNF、NT-3 m RNA及MBP蛋白的表达与模型组比较均明显上调(P<0.05)。LFB髓鞘染色显示治疗组髓鞘结构较模型组排列紧密,空泡减少,具有显著性差异(P<0.05)。结论:神经根磁刺激可以促进慢性不完全性脊髓损伤大鼠髓鞘结构的修复。Objective:To observe the effect of nerve root magnetic stimulation on myelin repair in rats with chronic incomplete spinal cord injury.Method:Eighteen male SD rats were randomly divided into sham operation group,model group and treatment group,with 6 rats in each group.The rat model of spinal cord injury with vertical clamp was established.The treatment group was given therapy for 14 days from the 11 th day after spinal cord injury.The expression of brain-derived neurotrophic factor(BDNF),neurotrophin-3(NT-3)m RNA and glial fibrillary acidic protein(GFAP)in the injured spinal cord were detected by quantitative real-time PCR.The expression of MBP in the injured area was observed by Western blot and immunofluorescence staining.The pathological changes of myelin in the injured area were observed by LFB staining.Result:Quantitative real-time PCR showed that the expression of BDNF and NT-3 m RNA were significantly up-regulated(P<0.05).Western blot showed that the expression of MBP was higher than that of the model group.LFB staining showed that the myelin structure of the treatment group was more closely arranged than that of the model group accompanied by the decreased number of vacuoles(P<0.05).Conclusion:Nerve root magnetic stimulation can promote the repair of myelin in rats with chronic incomplete spinal cord injury.
