摘 要:目的:探讨间歇运动调控心肌梗死(myocardial infarction,MI)大鼠肾脏miR-155/SIRT1/FoxO3a通路抑制肾脏细胞凋亡的作用。方法:3月龄雄性SD大鼠,左冠脉前降支结扎制备心梗模型,术后随机分为假手术组(Sham)、心肌梗死组(MI)和心梗+间歇运动组(ME),每组12只,其中Sham组实施假手术,只穿线不结扎。术后1周,ME组采用大鼠跑台进行4周间歇训练干预。训练结束后次日取材,测定各组大鼠心电图变化,RT-qPCR检测肾脏SIRT1及肾脏和血液miR-155表达,Western blot检测肾脏SIRT1、FoxO3a、caspase-3、cleaved-caspase-3、Bax和Bcl-2蛋白表达。结果:与Sham组比较,MI组大鼠肾脏Bax蛋白表达增加,Bcl-2蛋白表达及Bcl-2/Bax比值显著降低,FoxO3a、caspase-3、cleaved-caspase-3蛋白均显著增加,肾脏及血液miR-155表达均显著增加,SIRT1 mRNA和蛋白表达显著降低;与MI组比较,ME组大鼠肾脏Bax蛋白表达减少,Bcl-2蛋白表达增加,Bcl-2/Bax比值升高,FoxO3a、caspase-3、cleaved-caspase-3蛋白均显著降低,肾脏及血液miR-155表达均显著降低,SIRT1 mRNA和蛋白表达显著增加。肾脏SIRT1蛋白表达与FoxO3a、cleaved-caspase-3和Bax蛋白表达呈显著负相关,与Bcl-2蛋白表达呈显著正相关;肾脏miR-155表达与SIRT1蛋白表达呈显著负相关,与FoxO3a、cleaved-caspase-3和Bax蛋白表达呈显著正相关,与Bcl-2蛋白表达呈显著负相关。结论:间歇运动抑制miR-155表达,激活其下游SIRT1/FoxO3a信号通路。表明miR-155/SIRT1/FoxO3a信号通路在间歇运动抑制心梗大鼠肾脏细胞凋亡中发挥重要作用。Objective:The present study was to determine the effects of aerobic interval training(AIT)on the expressions of miR-155,SIRT1 and apoptosis in the kidney of rats with myocardial infarction(MI).Methods:male SD rats were randomly divided into:sham-operated group(Sham),sedentary MI group(MI)and MI with AIT group(ME)(n=12).The MI model was established by ligation the left anterior descending coronary artery.Rats in Sham groups were subjected to the same surgery,but only threaded and not ligated.After surgery 1 week,rats in ME groups took adaptability training and then subjected to 4 weeks treadmill exercise training.After training,the expression of renal SIRT1 mRNA and renal and serum miR-155 was examined by RT-qPCR.The expression of renal SIRT1,Bax,Bcl-2,FoxO3a,Caspase-3 and cleaved-Caspase-3 protein was examined by western blotting.Results:Compared with the Sham group,the expression of renal Bax protein was increased in MI group,the expression of Bcl-2 protein and the ratio of Bcl-2/Bax were significantly reduced in MI group,and the expression of FoxO3a,caspase-3 and cleaved-caspase-3 protein was augmented in the kidney of rats with MI.Moreover,the expression of SIRT1 mRNA and protein was obviously decreased in MI group.The level of renal and serum miR-155 was significantly increased in MI group.Furthermore,compared with the MI group,AIT increased the expression of SIRT1 mRNA and protein and reduced the level of renal and serum miR-155 in the kidney of ME group;Meanwhile,the expression of FoxO3a,caspase-3,cleaved-caspase-3 and Bax protein were diminished,and the expression of Bcl-2 protein and the ratio of Bcl-2/Bax were significantly increased in the ME group.The expression of SIRT1 protein was negatively related to the expression of FoxO3a,cleaved-caspase-3 and Bax protein,and positively related to the expression of Bcl-2 protein.The expression of miR-155 was negatively related to the expression of SIRT1 and Bcl-2 protein,and positively related to the expression of FoxO3a,cleaved-caspase-3 and Bax protein.Conclus
