摘 要:目的观察甘草苷对脑卒中后抑郁(PSD)大鼠额前皮质脑源性神经营养因子(BDNF)及凋亡因子Bax、Bcl-2蛋白表达的影响。方法随机选择SD大鼠10只为正常对照组。另50只SD大鼠用线栓法建立局灶性脑缺血大鼠模型后,随机分为脑卒中组、PSD组、甘草苷组、西酞普兰组和生理盐水组,每组10只,后4组大鼠再建立PSD模型。造模第43天时,各组大鼠额前皮质取材,用免疫荧光染色测大鼠BDNF和Bax、Bcl-2阳性细胞表达,用Western blot测各组BDNF和Bax、Bcl-2蛋白表达。结果各组大鼠额前皮质均可见BDNF、Bax及Bcl-2发红色荧光的阳性细胞表达。与正常对照组比较,PSD组大鼠BDNF及Bcl-2吸光度(A)值明显降低,Bax A值明显升高(P<0.05),与甘草苷组比较,PSD组、脑卒中组及生理盐水组大鼠BDNF、Bcl-2 A值明显降低,Bax A值升高(P<0.05),与西酞普兰组比较,PSD组大鼠BDNF、Bcl-2 A值明显降低(P<0.05),Bax A值明显升高(P<0.05)。与正常对照组比较,PSD组大鼠BDNF、Bcl-2蛋白表达明显减少(P<0.05),Bax蛋白表达明显增多(P<0.05),与甘草苷组比较,PSD组、脑卒中组及生理盐水组大鼠BDNF、Bcl-2蛋白表达明显减少,Bax蛋白明显增多(P<0.05),与西酞普兰组比较,PSD组大鼠BDNF(0.151±0.013 vs 0.252±0.013)、Bcl-2(0.199±0.057 vs 0.338±0.062)蛋白表达明显减少,Bax蛋白(0.448±0.023 vs 0.358±0.012)明显增多(P<0.05)。结论中药甘草苷可能有减少PSD大鼠额前皮质促凋亡因子Bax表达和(或)促进抗凋亡因子Bcl-2及BDNF表达的作用。Objective To study the effect of liquiritin on expressions of brain-derived neurotrophic factor(BDNF)and apoptosis factors,Bax and Bcl-2,in prefrontal cortex of poststroke depression(PSD)rats.Methods Ten SD rats served as a normal control group and 50 rats were randomly divided into stroke group,PSD group,liquiritin group,citalopram group and normal saline(NS)group(10 in each group).A focal cerebral ischemic model of rats was established by Longa occlusion.A PSD model was established for the latter 4 groups.The prefrontal cortex tissue samples were taken on day 43 after the model was established.The expressions of BDNF,Bax and Bcl-2 positive cells were detected with immunofluorescence staining and those of BDNF,Bax and Bcl-2 proteins were detected by Western blot.Results The expressions of BDNF,Bax and Bcl-2 positive cells were detected in rat prefrontal cortex of different groups.The average absorbance of BDNF and Bcl-2 was significantly lower while that of Bax was significantly higher in PSD group than in normal control group(P<0.05),in PSD group,stroke group and NS group than in liquiritin group(P<0.05)and in PSD group than in citalopram group(P<0.05).The expression level of BDNF and Bcl-2 was significantly lower while that of Bax was significantly higher in PSD group than in normal control group(P<0.05),in PSD group,stroke group,NS group than in liquiritin group(P<0.05),and in PSD group than in citalopram group(0.151±0.013 vs 0.252±0.013,0.199±0.057 vs 0.338±0.062,P<0.05;0.448±0.023 vs 0.358±0.012,P<0.05).Conclusion Liquiritin can downregulate the expression of Bax and upregulate the expression of Bcl-2 and BDNF in prefrontal cortex of PSD rats.
