摘 要:目的观察不同浓度大黄水提取物对化疗大鼠营养水平及肠道黏膜屏障功能的影响。方法将100只Wistar大鼠随机分为空白组、模型组及大黄水提取物低、中、高剂量组5组,各20只。除空白组外,其余4组大鼠采用注射用奥沙利铂+氟尿嘧啶注射液腹腔注射建立腹腔化疗模型。大黄水提取物低、中、高剂量组在建立化疗模型同时分别予0.5、1.0、2.0 g/kg大黄水提取物灌胃,连续给药6 d。空白组、模型组常规饲养。比较各组大鼠造模前后一般状态,造模前、给药后营养相关指标(体质量、摄食量和前白蛋白)变化情况;比较各组大鼠实验第7天血内毒素含量、回肠黏膜厚度、结肠黏膜厚度及肠系膜淋巴结和肝组织细菌移位阳性率。结果与本组造模前比较,造模后模型组体质量、摄食量、前白蛋白均降低(P<0.05),且低于空白组(P<0.05),大黄水提取物低、中剂量组体质量均降低(P<0.05),大黄水提取物低、中、高剂量组摄食量、前白蛋白均降低(P<0.05);与模型组比较,造模后大黄水提取物中、高剂量组体质量、摄食量、前白蛋白均增加(P<0.05),大黄水提取物高剂量组摄食量、前白蛋白较大黄水提取物中剂量组增加(P<0.05)。与空白组比较,模型组血内毒素含量升高(P<0.05),回肠黏膜厚度、结肠黏膜厚度降低(P<0.05);与模型组比较,大黄水提取物中、高剂量组血内毒素含量降低(P<0.05),回肠黏膜厚度、结肠黏膜厚度增高(P<0.05);与大黄水提取物中剂量组比较,大黄水提取物高剂量组血内毒素含量降低(P<0.05),回肠黏膜厚度、结肠黏膜厚度增高(P<0.05)。肠系膜淋巴结和肝组织细菌移位阳性率模型组高于空白组(P<0.05),大黄水提取物中、高剂量组均低于模型组(P<0.05),大黄水提取物低、中剂量组均高于大黄水提取物高剂量组(P<0.05),大黄水提取物低剂量组高于大黄水提取物中剂量组(P<0.05)。结论大黄水提Objective To investigate the effects of different concentrations of rhubarb water extract(RWE)on nutritional level and intestinal mucosal barrier function in chemotherapy rats.Methods Totally 100 Wistar rats were selected and randomly divided into 5 groups(control group,model group,low-dose group,medium-dose group,and high-dose group)with 20 rats each.Except for the blank group,rats in the other four groups were injected with oxaliplatin and 5-fluorouracil to establish intraperitoneal chemotherapy models.The same time,0.5,1.0 and 2.0 g/kg rhubarb water extract were additionally given in low-dose group,medium-dose group,and high-dose group for 6 days,respectively;the blank group and model group were routine feeded.This data were compared among the groups,such as the general status,nutrition-related indicators,blood endotoxin level,the thickness of the ileum mucosa,the thickness of the colonic mucosa,the changes in the positive rate of bacterial translocation in mesenteric lymph nodes and liver tissues.Results After modeling,body weight,food intake and prealbumin in the model group were lower than those before modeling,which were lower in the model group than those in the blank group and the medium-dose and the high-dose group(all P<0.05);the body weight in low-dose and medium-dose groups was lower than that before modeling(P<0.05);the food intake and prealbumin in all dose groups were lower than those before modeling(P<0.05).The food intake and prealbumin in the high-dose group were higher than those in the medium-dose group(P<0.05).Comparison of blood endotoxin content between all groups,the model group>the blank group,the model group>the medium-dose group>the high-dose group.Compared the thickness of ileum mucosa and colon mucosa between all groups,the model group<the blank group,the model group<the medium-dose group<the high-dose group.Compared the positive rate of bacterial translocation in mesenteric lymph nodes and liver tissues,the model group>the blank group,the model group>the medium-dose and high-dose gr
