摘  要:WRKY蛋白是植物中最大的转录因子家族之一,其家族成员广泛参与植物生长发育,特别在应答环境逆境胁迫中起重要的调节作用。开展WRKY家族成员在应答各种逆境胁迫中的作用及其机制研究,有利于阐明植物应答逆境胁迫的分子机制。本研究采用染色体步移技术,分离获得PsWRKY22的启动子,通过启动子5’端缺失体构建、拟南芥遗传转化和GUS酶活测定等方法研究pPsWRKY22及其缺失体在不同胁迫下(干旱,低温和高盐)以及外源激素(ABA,MeJA,SA和ETH)处理下的表达情况。研究结果表明,PsWRKY22启动子及其缺失片段在多种胁迫处理下都不同程度地抑制下游GUS蛋白的表达,尤其在外源SA胁迫处理下两者被抑制的效果最为明显,另外在低温和外源ABA处理下,pPsWRKY22及其缺失体的GUS酶活差异极其显著。本研究结果为进一步探究WRKY转录因子在非生物胁迫和外源激素作用下诱导油柰抗逆性基因表达的分子机制提供理论依据。WRKY proteins whose family members are considered to participate in the developmental process,are one of the largest transcription factor families in the plants,especially WRKY proteins have important roles in responses to biotic and abiotic stresses.The exploring roles and responding mechanisms of WRKY family members under various stresses would be beneficial to clarify molecular mechanism of responses under different conditions of stresses in plants.The promoter of PsWRKY22 was isolated by Genomic Walking technology.Expression of pPsWRKY22 and its deletion fragment under different stress conditions(drought,low temperature and high salinity)and exogenous hormones(ABA,MeJA,SA and ETH)by construction of 5’-terminal deleted fragments of the promoter,genetic transformation of Arabidopsis thaliana and GUS activity assay were detected.The results showed that the downstream GUS activity driven by PsWRKY22 promoter and its deleted fragments were inhibited under various stress treatments,especially for the exogenous SA treatment.In addition,the GUS activity of pPsWRKY22 and its deletions were significantly changed under low temperature and ABA treatments,which would lay solid foundation for finding the molecular mechanism of WRKY transcription factors in responses to exogenous hormones and abiotic stresses.
