摘 要:叶酸是一类水溶性B族维生素,它是人体生命活动所必需的微量营养物质。由于人体不能自身合成叶酸,只能依靠食物摄取。因此利用遗传育种手段培育高叶酸含量的鲜食甜、糯玉米品种是人类叶酸营养强化的重要途经。玉米谷氨酸亚胺甲基转移酶Ⅰ基因(ZmGFT1)是叶酸含量相关的关键基因,普通玉米该等位基因的SNP位点导致天冬酰胺转变为甘氨酸,从而影响总叶酸的含量。2个位点处的碱基均为G的玉米叶酸含量显著高于2个位点处的碱基均为A的玉米叶酸含量。本研究通过检测218份甜、糯玉米自交系GFT1基因的功能SNP位点发现该位点存在3种类型的等位变异,即GGG、CAA、GAA,根据功能SNP位点设计CAPS标记GFT1-2。利用该标记对甜玉米和糯玉米GFT1基因型进行鉴定,与测序结果一致。同时检测了鲜果穗叶酸含量,结果表明GFT1-2标记能准确鉴定GFT1的基因型,可用于加快高叶酸含量甜玉米和糯玉米的分子标记辅助育种。Folates are a type of water-soluble B vitamins,which is a micronutrient necessary for human.Because human cannot synthesize folates de novo,it can only rely on food intake.Therefore,the use of genetic breeding methods to cultivate high-folate sweet and waxy corn varieties is an important way for human.Maize glutamate formiminotransferaseⅠgene(ZmGFT1)is a key gene related to the content of folates.The SNP site of the GFT1 allele of maize causes the change of asparagine to glycine,which affects the content of folates.The content of folates with bases G at both sites was significantly higher than that with bases A at both sites in normal maize.In this study,we have detected functional SNP loci of GFT1 gene in 218 sweet and waxy maize inbred line three types of alleles,namely GGG,CAA,and GAA,were found in this locus.The CAPS marker GFT1-2 was designed based on the functional SNP locus.The CAPS marker was used to identify GFT1 genotypes of sweet corn and waxy corn,which was consistent with the sequencing results.The folate contents of fresh kernel were also tested,and the results showed that the GFT1-2 marker could accurately identify the genotype of GFT1 and could be used to accelerate molecular marker-assisted breeding of sweet corn and waxy corn with high folates.
