露蕊乌头产C_(20)型二萜生物碱细胞系的建立

摘　　要：为建立和明确露蕊乌头愈伤组织的诱导体系,本研究以露蕊乌头无菌苗的根为外植体,以诱导率为愈伤组织诱导情况的指标筛选最佳植物激素配比,并观察不同外植体诱导愈伤组织的能力和细胞系的增殖情况;此外,通过超高效液相色谱-四级杆-飞行时间质谱(UPLC-Q-TOF-MS)鉴定细胞系中的成分组成。结果表明:露蕊乌头诱导愈伤组织的最佳培养基为MS+3.0 mg/L 6-BA+2.0 mg/L 2,4-D,愈伤组织的诱导能力为根>叶>种子,30 d后愈伤组织细胞系的增殖率均达到3500%以上。细胞系中的主要成分为C_(20)型二萜生物碱阿替生C_(22)H_(33)NO_(2),含量达到200μg/g以上。本研究建立了露蕊乌头愈伤组织的诱导体系,为C_(20)型生物碱的开发和应用提供参考。In order to establish and define the callus induction system for Aconitum gymnandrum Maxim.,the optimal plant hormone ratio was selected based on the callus induction rate and the callus induction ability of different explants and the proliferation of cell lines were observed.In addition,the components in the cell lines were identified by UPLC-Q-TOF-MS.The results showed that the best medium for callus induction was MS+3.0 mg/L 6-BA+2.0 mg/L 2,4-D.The callus induction ability was root>leaf>seed.After 30 days,the proliferation rate of callus cell lines reached more than 3500%.C_(20)diterpene alkaloid atisine was the main component in the cell lines of A.gymnandrum,the content of which reached more than 200μg/g.In this study,a callus induction system was established for the development and application of C_(20)alkaloids.
