摘  要:为了筛选出适合百蕊草阶段式培养的快繁方案,建立较为完善的百蕊草植株再生体系,为后期人工栽培和工厂化育苗提供技术支持。本研究以百蕊草(Thesium chinense Turcz)的茎、叶为外植体,研究不同浓度和种类的激素配比以及外源添加物组合对百蕊草阶段式培养的影响。结果显示,百蕊草愈伤组织诱导的最佳培养基为MS+0.2 mg/L萘乙酸(NAA)+1.0 mg/L 6-苄氨基嘌呤(6-BA)+0.2 mg/L 2,4-二氯苯氧乙酸(2,4-D);诱导丛芽的最佳培养基为MS+1.5 mg/L噻苯隆(TDZ);繁殖阶段的最佳培养基为MS+2.0 mg/L NAA+0.5 mg/L6-BA+10 mg/L椰子汁;诱导瓶内生根的最佳培养基为1/2MS+2.0 mg/L NAA+0.5 mg/L 6-BA+0.3 mg/L ABT。使用瓶外生根的方法时,百蕊草外植体用60 mg/L的ABT生根粉溶液浸泡1 h后诱导生根的效果最好。本研究优选的百蕊草离体培养和植株再生体系稳定可靠,可为后期的开发利用提供有力的技术支持。In order to screen out the rapid propagation scheme suitable for the stage cultivation of Thesium chinense Turcz,and establish a relatively perfect plant regeneration system,so as to provide technical support for the later artificial cultivation and factory nursery.In this study,the stems and leaves of Thesium chinense Turcz were used as explants to study the effects of different concentrations and types of hormone ratios and the combination of exogenous additives on the stage cultivation of Thesium chinense Turcz.The results showed that the optimum medium for the induction of callus of Thesium chinense Turcz is MS+0.2 mg/L naphthaleneacetic acid(NAA)+1.0 mg/L 6-benzylaminopurine(6-BA)+0.2 mg/L 2,4-dichlorophenoxyacetic acid(2,4-D);the optimum medium for the induction of bud is MS+1.5 mg/L thidiazem(TDZ);the optimum medium for reproduction stage is MS+2.0 mg/L NAA+0.5 mg/L 6-BA+10 mg/L coconut juice;the optimum medium for inducing rooting in the bottle is1/2 MS+2.0 mg/L NAA+0.5 mg/L 6-BA+0.3 mg/L ABT.When using the method of rooting outside the bottle,the effect of rooting induced by the root explants of Thesium chinense Turcz was soaked in 60 mg/L ABT rooting water for 1 h.The preferred in vitro culture and plant regeneration system of Thesium chinense Turcz is stable and reliable,which can provide strong technical support for later development and utilization.
