摘 要:目的:通过高脂饮食诱导复制非酒精性脂肪性肝炎大鼠模型,观察祛脂愈肝汤对非酒精性脂肪性肝炎大鼠血清IL-17、IL-2表达的影响,探索祛脂愈肝汤干预NASH的作用机制。方法:通过高脂饮食诱导NASH大鼠模型,经病理学验证动物模型后,将高脂饮食组大鼠随机分为模型对照组,多烯磷脂酰胆碱组,祛脂愈肝汤低、中、高剂量组。给予祛脂愈肝汤进行灌胃治疗,采用多烯磷脂酰胆碱进行药效比较。未诱导的大鼠作为空白对照。8周后处死大鼠检测血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、甘油三酯(TG)、总胆固醇(TC)水平,酶联免疫吸附测定法(ELISA)检测血清白细胞介素17(IL-17)、白细胞介素2(IL-2)水平。结果:与空白组相比,模型对照组大鼠血清AST、ALT、TG、TC水平显著升高(P<0.01);血清IL-17显著升高,IL-2显著降低(P<0.01);药物干预组与模型对照组相比,治疗8周后血清AST、ALT、TG、TC水平、IL-17水平显著降低(P<0.01),IL-2水平显著升高(P<0.01);其中祛脂愈肝汤高剂量组与多烯磷脂酰胆碱的作用差异无统计学意义(P>0.05)。结论:祛脂愈肝汤对非酒精性脂肪性肝炎具有良好的治疗作用,其作用机制可能与调控IL-17、IL-2表达有关。Objective:The study aimed to evaluate the effects of Quzhi Yugan Decoction on serum IL-17 and IL-2 levels in liver tissues of non-alcoholic steatohepatitis(NASH)model rats.The NASH model rats induced by high-fat diet,and the intervention mechanism of Quzhi Yugan Decoction on NASH in rats were studied.Method:High-fat diet was used to establish a rat model of NASH.The non-alcoholic steatohepatitis levels were assessed by using pathological examination,the rats in the high-fat diet group were randomly divided into the model control group,the Polyene Phosphatidylcholine group,and the low-dose,medium-dose and high-dose Quzhi Yugan Decoction groups.Untreated rats were as background group.Quzhi Yugan Decoction was administered by gavage,and Polyene Phosphatidylcholine was used to compare its efficacy.After 8 weeks,the rats were sacrificed to detect the levels of AST,ALT,triglyceride(TG),total cholesterol(TC)in the plasma.In addition,serum levels of interleukin 17(IL-17)and interleukin 2(IL-2)were determined by ELISA.Result:Compared with the background group,serum levels of AST,ALT,TG and TC in the model control group were significantly increased(P<0.01),level of serum IL-17 was significantly increased and serum IL-2 was significantly decreased(P<0.01).Compared with the model control group,serum AST,ALT,TG and TC levels,IL-17 content were significantly decreased in the drug intervention group after 8 weeks of treatment(P<0.01),and IL-2 content was significantly increased(P<0.01).There was no significant difference between Quzhi Yugan Decoction high dose group and Polyene Phosphatidylcholine(P>0.05).Conclusion:Quzhi Yugan Decoction showed good therapeutic effects on NASH,and its mechanism may be related to the regulation of the expression of IL-17 and IL-2.
