摘 要:目的通过重组腺病毒介导使低氧诱导因子-2α(HIF-2α)过表达,探讨其过表达对SD孕鼠的影响。方法将SD孕鼠随机分为重组腺病毒组、盐水组、空白腺病毒组,每组12只,于妊娠第9 d经鼠尾静脉分别注射100μL载有HIF-2α的重组腺病毒(1×10^(10)Pfu/只)、盐水、空白腺病毒(1×10^(10)Pfu/只),观测孕鼠血压、尿蛋白、肝肾功能情况及仔鼠均重变化,应用免疫组化法分析胎盘组织HIF-2α表达情况。结果妊娠第19 d,与空白腺病毒组、盐水组相比,重组腺病毒组血压、尿蛋白显著升高,仔鼠均重明显降低,差异具有统计学意义(P<0.05)。空白腺病毒组与盐水组相比,孕鼠血压、尿蛋白、仔鼠均重无明显差异(P>0.05)。三组孕鼠ALT、ALB、BUN、Cr无明显差异(P>0.05),免疫组化结果提示重组腺病毒组HIF-2α表达高于盐水组、空白腺病毒组(P<0.05)。结论 HIF-2α过表达可使SD孕鼠产生子痫前期症状、胎儿宫内生长受限(IUGR),HIF-2α可能参与子痫前期发病机制。Objective To investigate the effects of hypoxia inducible factor-2α(HIF-2α)overexpression mediated by recombinant adenovirus on pregnant SD rats.Methods Pregnant SD rats were randomly divided into recombinant adenovirus group, saline group and blank adenovirus group with 12 rats in each group.On the 9 th day of pregnancy, 100 μL recombinant adenovirus carrying HIF-2α(1×10^(10)PFU/rat),saline group and blank adenovirus(1×10^(10)PFU/rat)were injected into tail vein in rats to observe the blood pressure, urine protein, liver and kidney function of pregnant rats and average weight of offspring.In addition, the expression of HIF-2α in placenta was analyzed by immunohistochemistry.Results On the 19 th day of pregnancy, compared with the blank adenovirus group and saline group, the blood pressure and urine protein of recombinant adenovirus group were significantly increased, and the average weight of offspring was significantly decreased(P<0.05).The difference was statistically significant(P<0.05).There was no significant difference in blood pressure, urine protein and weight of offspring between blank adenovirus group and saline group(P>0.05).There was no significant difference in ALT,ALB,BUN and Cr among the rats in three groups(P>0.05).The results of immunohistochemical showed that the expression of HIF-2α in recombinant adenovirus group was higher than that in saline group and blank adenovirus group(P<0.05).Conclusion Overexpression of HIF-2α can induce preeclampsia symptoms and fetal growth restriction in utero(IUGR)in pregnant SD rats, and HIF-2α may be involved in the pathogenesis of preeclampsia.
