摘  要:本研究以温州芜菁(Brassica rapa L. em. Matzg.,2n=20,AA)为母本,水果紫苤蓝(B. oleracea L. var. caulorapa DC,2n=18,CC)为父本进行远缘杂交,创造种间杂种。通过花期剥蕾授粉、子房培养共获得2株杂种,经扩繁得到无性苗19株,经形态学分析、细胞学观察和RAPD分子鉴定出2株均为温州芜菁×紫苤蓝的真杂种,后利用染色体加倍获得异源四倍体新型芜菁甘蓝新种质材料(AACC,2n=38)。两杂种株系出现茎色分离,根部与茎部均有膨大,但程度低于亲本。异源四倍体的花器官较大,花药发育正常,可育花粉率高。本研究所创制的异源四倍体新种质,在一定程度上拓展了芸薹属蔬菜种质资源的遗传基础。In the present study,wide hybridization was performed between Whenzhou turnip(Brassica rapa L. em. Matzg.,2 n=20,AA)and purple kohlrabi(B. oleracea L. var. caulorapa DC,2 n=18,CC)using hand emasculation,followed by the tissue culture for hybrid seeds. A total of two hybrids were obtained through bud pollination and ovary culture. Morphological analysis,cytological observation and RAPD molecular analysis further confirmed the two strains as hybrids of Wenzhou turnip × purple kohlrabi. Nineteen asexual seedlings were obtained through expanding propagation,and the novel allotetraploid rutabagas germplasm was obtained by chromosome doubling(AACC,2 n=38). The asexual plants derived from two hybrids were different on stem color,and enlarged roots and stems,lower than those of the parents. The allotetraploids represented larger floral organs,higher pollen viabilities and anthers developed as expected. The new allotetraploid generated from this study provided possibility on enriching the genetic diversity of Brassica germplasm resources.
