摘 要:目的探讨姜黄素对糖尿病足溃疡小鼠氧化应激、细胞炎症因子和创面血管新生的影响。方法选择SPF级雄性C57小鼠(BKS背景)60只,随机分为对照组、模型组和姜黄素组,每组20只。高脂高糖喂养联合腹腔内注射链脲佐菌素(STZ)建立糖尿病模型,模型组造模成功18只,姜黄素组造模成功17只,3组随机取15只小鼠进行相关研究。姜黄素组给予姜黄素100 mg/kg灌胃,每日1次;对照组和模型组小鼠给予等剂量生理盐水灌胃,每日1次。比较3组小鼠氧化应激、细胞炎症因子、创面血管新生和创面愈合情况。结果与对照组比较,模型组和姜黄素组血浆MDA水平升高,而SOD活性下降(P<0.05);姜黄素组血浆MDA水平低于模型组,而SOD活性高于模型组(P<0.05)。与对照组比较,模型组和姜黄素组血浆IL-1和TNF-α水平升高(P<0.05);姜黄素组血浆IL-1和TNF-α水平低于模型组(P<0.05)。与对照组比较,模型组和姜黄素组新生毛细血管数密度降低(P<0.05);姜黄素组新生毛细血管数密度高于模型组(P<0.05)。与对照组比较,模型组和姜黄素组创面愈合率降低(P<0.05);姜黄素组创面愈合率高于模型组(P<0.05)。结论姜黄素对糖尿病足溃疡小鼠可促进创面血管新生和创面愈合,减轻炎症反应,且具有抗氧化应激作用。Objective To investigate the effects of curcumin on oxidative stress,inflammatory cytokines and wound angiogenesis in diabetic foot ulcer mice.Methods A total of 60 male C57 mice(BKS background)of SPF grade were randomly divided into control group(n=20),model group(n=20)and curcumin group(n=20).A diabetic rat model was established by intraperitoncal injection with streptozotocin and breeding with high fat and glucose.A diabetic rat model was established by intraperitoncal injection with streptozotocin(STZ)and breeding with high fat and glucose.Among them,18 mice in the model group and 15 mice in the curcumin group were successfully modeled.And 15 mice from each group were selected for study.Mice in the curcumin group were given curcumin 100 mg/kg once a day by gavage,while mice in the control group and the model group were given the same dose of normal saline once a day.The oxidative stress,inflammatory cytokines,wound angiogenesis and wound healing in each group were compared.Results Compared with the control group,plasma MDA level in model group and curcumin group increased,while SOD activity decreased(P<0.05).Plasma MDA level in curcumin group was lower than that in model group,while SOD activity was higher than that in model group(P<0.05).Compared with the control group,plasma levels of IL-1 and TNF-αin model group and curcumin group increased(P<0.05).Plasma IL-1 and TNF-αlevels in curcumin group were lower than those in model group(P<0.05).Compared with the control group,the density of new capillaries in model group and curcumin group decreased(P<0.05).The density of new capillaries in curcumin group was higher than that in model group(P<0.05).ompared with the control group,the wound healing rate of model group and curcumin group was lower(P<0.05).The wound healing rate of curcumin group was higher than that of model group(P<0.05).Conclusion Treating diabetic foot ulcer mice with curcumin can promote wound angiogenesis and wound healing,reduce inflammation,and has anti-oxidative stress effect.
