摘  要:用人工饲料饲养的家蚕存在生长发育状况差、产丝量低、造卵数少、活力弱等问题。为解析造成这些问题的分子机制,采用高通量测序的方法,比较桑叶育和人工饲料育家蚕化蛹第3天雌雄蛹的转录组差异。结果显示,与人工饲料育家蚕相比,桑叶育家蚕雌蛹中共鉴定出61个差异表达基因,其中45个基因上调表达,16个基因下调表达;雄蛹中共鉴定出227个差异表达基因,其中134个基因上调表达,93个基因下调表达。结合GO分析和KEGG通路分析,发现雌蛹中差异表达基因显著富集在细胞色素P450对外源物质代谢的通路,而雄蛹中差异表达基因主要富集在糖胺聚糖降解、糖酵解和糖异生等途径。人工饲料喂养组中雌蛹的谷胱甘肽-S-转移酶基因表达显著下调,可能影响雌蚕体内激素的合成,从而影响生殖;而雄蛹中丙酮酸激酶基因的表达显著下调,可能影响正常的能量代谢,从而导致雄蛾活力不高。To clarify the molecular mechanism that results in poor growth,low silk production,low egg production and weak vitality of silkworms reared on artificial diet,the transcriptome of 3 rd-day pupa of female and male silkworms reared on mulberry leaf and artificial diet were analyzed with high-throughput sequencing.A total of 61 differentially expressed genes were identified in female pupa of mulberry leaf group,among which 45 differentially expressed genes were up-regulated and 16 genes were down-regulated.A total of 227 differentially expressed genes were identified in male pupa of mulberry leaf group,with 134 up-regulated genes and 93 down-regulated genes.Combined with GO analysis and KEGG pathway analysis,it was found that the differentially expressed genes in female pupa were significantly enriched in the cytochrome P450 pathway for metabolism of exogenous substances,while the differentially expressed genes in male pupa were mainly enriched in the pathways of glycosaminoglycan degradation,glycolysis and gluconeogenesis.Futhermore,the results showed that the glutathione-S-transferase gene in female pupa of artificial diet group was significantly down-regulated,which may affect the hormone synthesis and reproduction in female silkworm.In addition,the pyruvate kinase gene in male pupa of artificial diet group was significantly down-regulated,which may affect the normal sugar metabolism,resulting in weak vitality of male moths.
