摘 要:目的:研究肉桂通过调控低氧诱导因子1α(HIF-1α)表达,对慢性缺氧心肌细胞的保护作用。方法:将心肌细胞H9c2分为空白对照组、缺氧组、肉桂低剂量组、肉桂中剂量组、肉桂高剂量组。CCK-8法检测各组细胞增殖情况,TUNEL法检测各组细胞凋亡情况。应用试剂盒检测各组细胞超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量和硫代巴比妥酸反应产物(TBARS)水平;Western blot检测心肌肌钙蛋白T(cTnT)、肌酸激酶同工酶MB(CK-MB)、肌红蛋白(Myo)、HIF-1α和血管内皮因子(VEGF)表达。结果:缺氧处理后显著降低了心肌细胞H9c2增殖活力、SOD活性,且增加了细胞凋亡指数、MDA和TBARS含量(均P<0.05)。缺氧处理后显著降低了心肌细胞H9c2的cTnT、CK-MB、Myo、HIF-1α和VEGF表达量(均P<0.05)。各剂量肉桂组能够不同程度的逆转缺氧对心肌细胞H9c2上述指标的影响。结论:肉桂通过调控HIF-1α表达保护缺氧诱导的大鼠心肌细胞损伤。Objective:To investigate the protective effect of cinnamon on chronic hypoxia cardiomyocytes by regulating the expression of hypoxia-inducible factor 1α(HIF-1α).Methods:Cardiomyocyte H9c2 were divided into blank control group,hypoxia group,cinnamon low-dose group,cinnamon medium-dose group and cinnamon high-dose group.Cell proliferation was determined by CCK-8 assay.Cell apoptosis was detected by TUNEL staining.The superoxide dismutase(SOD)activity,malondialdehyde(MDA)content and thiobarbituric acid reactive substances(TBARS)level were measured by kits.Expression of cardiac troponin T(cTnT),creatine kinase MB(CK-MB),myoglobin(Myo),HIF-1αand VEGF proteins were determined by Western blot.Results:Hypoxia treatment significantly reduced the cardiomyocyte H9c2 of SOD activity and increased apoptosis index,MDA and TBARS content(all P<0.05).After hypoxia treatment,cardiomyocyte H9c2 expression levels of cTnT,CK-MB,Myo,HIF-1 and VEGF were decreased(all P<0.05).Each dose of cinnamon group could reverse the effects of hypoxia on the above indexes of cardiomyocyte H9C2 to different degrees.Conclusion:Cinnamon protects hypoxia-induced cardiomyocyte injury in rats by regulating HIF-1αexpression.
