小麦叶片可溶性糖含量测定方法的研究

摘　　要：研究并完善了蒽酮硫酸法,即取可溶性糖溶液1mL,加蒽酮稀硫酸5mL,沸水浴10min,计时,第20min,于625nm测吸光度值。吸光度值超过0.8应先稀释该溶液。做标准曲线,即取10~100μg/mL的葡萄糖溶液1mL,其他方法同上。小麦叶片可溶性糖的提取,称干叶片0.01g,剪碎,放入离心管,加10mL蒸馏水,沸水浴60min,加30mg活性炭,混匀,静止30min,过滤,加3mL无水乙醇,蒸馏水定容至15mL。First studied and improved the anthrone sulphuric acid method,the soluble sugar extracted solution 1 mL in the scale centrifuge tube,add anthrone dilute sulphuric acid solution 5 mL,twist the lid,reaction time of 10 min by boiling water bath,then start timing,centrifugal shell ice water bath or tap water rinse to room temperature,20 min,use ultraviolet spectrophotometer wavelengths of 625 nm absorbance measurement values.If the absorbance value exceeds 0.8,the soluble sugar extract should be diluted first.The standard curve was made with glucose solution,namely 1mL of glucose solution with a concentration of 10-100ug/mL was used in the scale centrifugal tube.Other methods were the same as above.Method for extracting soluble sugar in wheat leaves:0.01g dry leaves,cut into pieces and put them into 15mL scale centrifugation,add 10mL distilled water,twist the lid,boil water bath for 60min,cool them to room temperature,add 30mg activated carbon,mix them,rest them for 30min,filter them,add 3mL anhydrous ethanol,mix them with distilled water to 15mL,then mix them to get soluble sugar extract.
