摘 要:为建立一种理想的急性肺损伤模型,筛选出小鼠急性肺损伤模型最佳造模时间和脂多糖浓度,本试验采用小鼠滴鼻给药途径,选取48只雄性SPF级小鼠随机分成8组,空白对照组、造模时间组(3、6、9 h和12 h)和造模剂量组[1、2 mg/(kg·bw)和5 mg/(kg·bw)]。分别检测各组小鼠肺湿干重比(W/D)、肺脏病理损伤、细胞因子(IL-6、IL-1β、TNF-α)含量、中性粒细胞百分比等指标,筛选出最佳给药剂量及时间。结果表明,脂多糖浓度为2 mg/(kg·bw)、造模时间6 h时,小鼠急性肺损伤模型更加稳定。In order to establish an ideal model of acute lung injury,the optimal model time and lipopolysaccharide concentration were selected.In this experiment,mice were administered by nasal drops,48 male SPF mice were selected and randomly divided into eight groups,blank control group,mold time group(3,6,9 h and 12 h)and mold dose group[1,2 mg/(kg·bw)and 5 mg/(kg·bw)].Lung wet and dry weight ratio(W/D),lung pathological sections,cytokines(IL-6,IL-1β,TNF-α)content and neutrophilic granulocyte percentage in each group were detected,and the optimal dose and time of administration were evaluated.The results showed that when lipopolysaccharide concentration was 2 mg/(kg·bw)and the modeling time was 6 h,the model of acute lung injury in mice was more stable.
