摘 要:为了建立丁香微乳的质量控制标准,用薄层色谱法对制剂中丁香进行定性鉴别,采用高效液相色谱法(HLPC)测定丁香酚含量,色谱柱为Welchrom C18(4.6 nm×250 nm,5μm),流动相为甲醇-水(55∶45),检测波长为282 nm。结果显示,丁香微乳薄层色谱中,在与对照药材或丁香酚对照品相应的位置上,显示相同颜色的荧光斑点,阴性对照无干扰;HPLC测定丁香酚含量的线性方程为y=22545602.5974 x+30256.8000,丁香酚在0.0110~0.0660 mg/mL范围内与峰面积有良好线性关系(r=1.0000),平均回收率为100.60%,RSD为0.99%。结果表明该方法简便易行,专属性强,重复性好,可用于丁香微乳的质量控制。To establish a quality control standard for Caryophylli Flos microemulsionThin layer chromatography(TLC)method was performed to identify eugenol in Caryophylli Flos microemulsion and the content of eugenol was determined by high performance liquid chromatography(HPLC).The column was Welchrom C18(46 nm×250 nm,5μm),the mobile phase was methanolwater(55∶45),and the detection wavelength was 282 nmFor the TLC of Caryophylli Flos microemulsion,fluorescent spots of the same color were shown at positions corresponding to the control drug or standard eugenol.The negative control had no interference.The linear equation of determined eugenol content by HPLC was y=225456025974x+302568000,and there was a linear relationship with the peak area in the range of 00110066 mg/mL(r=10000)The average recovery rate was 10060%,with RSD of 099%The result showed that the methods are simple and convenient with strong specificity and repeatability,which could be used for the quality control of Caryophylli Flos microemulsion。
