摘  要:目的:探讨女贞子治疗骨质疏松性骨折的作用成分及作用机制。基于网络药理学的理念,运用网络分析技术,预测女贞子治疗骨质疏松性骨折的潜在靶点并分析其作用机制。方法:利用TCMSP平台收集女贞子的中药活性成分,应用Perl软件预测潜在相关靶点信息,分别从GeneCards数据库、OMIM数据库中筛选出与骨质疏松性骨折相关联的靶点。将中药靶点与疾病靶点进行取交集,进而运用Cytoscape3.7.2构建中药调控网络,再采用STRING平台构建PPI网络,通过R语言对相关疾病-药物靶点基因进行GO富集分析和KEGG通路分析。结果:从女贞子中筛选出13个有效活性成分,包括槲皮素、木犀草素、山柰素、β-谷固醇、花旗松素等,与骨质疏松性骨折有关的作用靶点111个,主要有IL-6、AKT1、VEGFA、JUN、PTGS2和MAPK1等。涉及4条主要信号通路,包括TNF信号通路、PI3K/AKT信号通路、HIF-1信号通路和细胞凋亡等信号通路。这些通路可能与骨质疏松性骨折发生发展密切相关。结论:女贞子药物有效成分可能基于对TNF、PI3K/AKT、HIF-1信号通路的调控,发挥抑制炎症反应、促进破骨细胞凋亡等作用治疗骨质疏松性骨折。Objective:To investigate the functional components and mechanism of ligustrum lucidum in the treatment of osteoporotic fracture.Methods:TCMSP platform was used to collect active components of ligustrum lucidum,Perl software was used to predict the information of potential related targets,and targets associated with osteoporosis fracture were screened out from Genecards database and OMIM database,respectively.TCM targets were intersected with disease targets,and then the TCM regulatory network was constructed by Cytoscape3.7.2.Then the STRING platform was used to construct PPI network,and the GO enrichment analysis and KEGG pathway analysis of disease-drug target genes were carried out by R language.Results:Thirteen active components were screened from ligustrum ligustrum,including quercetin,luteolin,kaempferol,β-sterol,paclitaxel,etc.A total of 111 active targets related to osteoporotic fracture were identified,mainly including IL-6,AKT1,VEGFA,JUN,PTGS2 and MAPK1.Four major signaling pathways were involved,including TNF signaling pathway,PI3K/AKT signaling pathway,HIF-1 signaling pathway and apoptosis signaling pathway.These pathways may be closely related to the occurrence and development of osteoporotic fractures.Conclusion:The active components of ligustrum ligustrum may inhibit inflammatory response and promote osteoclast apoptosis in the treatment of osteoporotic fractures based on the regulation of TNF,PI3K/AKT and HIF-1 signaling pathways.
