摘 要:目的:建立从实验树鼩外周血中分离单个核细胞的可靠方法。方法:使用不同密度的Percoll®分离溶液对树鼩的静脉血进行密度梯度离心,并通过Wright-Giemsa染色和细胞计数分析树鼩的单核细胞的最佳分离密度。结果:采用60%和67%Percoll®分离溶液的不连续密度梯度离心方法可以分离出树鼩粒细胞。60%~63%的分离溶液可以成功地将静脉血分为4层,随着密度的增加,分离的细胞越多,粒细胞和红细胞受到污染的风险就越高。结论:在确保淋巴细胞纯度的前提下,60%Percoll®分离液可分离外周血中更多的单核细胞,可用于从树鼩静脉血中分离外周血单个核细胞。Objective:To establish a reliable method for the isolation of peripheral blood mononuclear cells from experimental tree shrews.Methods:The venous blood of tree shrews was subjected to density gradient centrifugation with Percoll®separation solutions of different densities,and the optimal separation density of the tree shrews mononuclear cells was analyzed by Wright-Giemsa staining and cell counting.Results:The discontinuous density gradient centrifugation method using 60%and 67%Percoll®separation solution could separate tree shrew granulocytes.60%-63%Percoll®separation solution could successfully separate the tree shrew venous blood into four layers,and the more the number of cells separated with the increase in density,the more serious the pollution of granulocytes and red blood cells.Conclusion:Under the premise of ensuring the purity of lymphocytes,60%Percoll®separation solution can separate more peripheral blood mononuclear cells,which can be used to separate peripheral blood mononuclear cells from tree shrews.
