摘 要:为了准确鉴定黑龙江省丹顶鹤体内的前殖吸虫并确定其分类地位,试验通过虫体染色、装片制作的方法对其进行形态学观察,采用PCR方法对该前殖吸虫的核糖体内转录间隔区(ITS)序列进行扩增、克隆和比较分析,并以ITS2序列为标记基因构建系统进化树。结果表明:丹顶鹤体内的前殖吸虫与透明前殖吸虫的形态学特点基本一致。该吸虫的ITS序列长度为1183 bp,其中ITS1、5.8S、ITS2的长度分别为852,107,224 bp,与国外报道的透明前殖吸虫(登录号为KP192734)同源性达到98%。丹顶鹤体内的透明前殖吸虫与国外已发表的透明前殖吸虫处于同一个分支,亲缘关系较近。说明丹顶鹤体内的吸虫为透明前殖吸虫,ITS2序列可以作为寄生虫虫种鉴定的良好基因标记。In order to accurately identify a species of Prosthogonimus spp.in Grus japonensis in Heilongjiang Province for the determination of its taxonomic status,the experiment was carried out through the method of dyeing the worm body to make morphological observations;the PCR method was used to amplify,clone and compare the internal ribosome transcribed spacer(ITS)sequence of the Prosthogonimus,and the ITS2 sequence was used as a marker gene to construct a phylogenetic tree.The results showed that the morphological characteristics of the Prosthogonimus in Grus japonensis were basically the same as that of Prosthogonimus pellucidus.The length of the ITS sequence of this fluke was 1183 bp,of which the lengths of ITS1,5.8 S,and ITS2 were 852,107,and 224 bp,respectively.The homology with Prosthogonimus pellucidus(accession number KP192734)reported abroad reached 98%.Prosthogonimus pellucidus in Grus japonensis was in a branch with Prosthogonimus pellucidus that had been published abroad,and was closely related.The results suggested that the fluke in Grus japonensis was Prosthogonimus pellucidus,and the ITS2 sequence can be used as a good genetic marker for the identification of parasite species.
