摘  要:试验旨在评价淫羊藿提取物的安全性,为临床用药提供参考依据。试验选取20只小鼠进行急性毒性试验,未检测出半数致死量(LD_(50)),另选取20只小鼠一次性灌服淫羊藿提取物(20 g/kg·BW)进行最大耐受量试验。将80只大鼠随机分为空白对照组,高、中、低剂量组(20、10、5 g/kg·BW),除空白对照组外,连续30 d灌服淫羊藿提取物并观察其变化。停药后继续观察7 d。观察大鼠体征和体重变化,采集血液进行血液生理生化指标测定,并进行病理组织学检查。结果显示,急性毒性试验各组小鼠均存活,体态无异常,未能测出LD_(50),小鼠对淫羊藿提取物的最大耐受量为20 g/kg·BW。30 d喂养试验中,各给药剂量组大鼠体重、血液生理生化指标和脏器指数与空白对照组相比,均无显著差异(P>0.05)。剖检心脏、肝脏、脾脏、肺脏、肾脏、胃肠、睾丸、卵巢和子宫等组织器官未见异常变化,镜检未见明显的病理组织学变化。结果表明,淫羊藿提取物属于无毒物质,对连续用药1个月的大鼠无毒性作用。The experiment was conducted to evaluate the safety of Epimedium extract and provide basis for clinical application. Twenty mice were selected for acute toxicity test, and the median lethal dose(LD50) was not detected. Another twenty mice were selected for maximum tolerance test, in which the animals were given Epimedium extract at a dose of 20 g/kg·BW each mice. 80 rats were randomly divided into blank control group and high, medium and low dose groups(20, 10, 5 g/kg·BW) and the rats were fed continuously with different dose Epimedium extract for 30 d except blank control group and observed for 7 d after drug withdrawal. The changes of physical signs and body weight of rats were observed during the administration. Blood of the rats was collected for measuring blood physiological and biochemical indexes. The pathological changes of the organs were observed and the indexes of the organs were measured. The results showed that the mice survived healthily without abnormal body sign in the acute toxicity test. The LD50 could not be determined and the maximum tolerance dose of mice to Epimedium extract was 20 g/kg·BW. In the 30-days feeding test,there was no significant differences in body weight, blood physiological and biochemical indexes, and organ index of rats in each administration group compared with the blank control group(P>0.05). No abnormal changes were observed in the heart, liver, spleen, lung, kidney, stomach and no obvious histopathological changes were observed under microscope. The results indicated that Epimedium extract was no acute toxicity to mice and also no subchronic toxicity to rats.
