摘 要:目的筛选出风寒咳喘方中的IL-1β调控活性成分并进行含量同时测定,以建立更为贴近药效机制的质量评价方法。方法根据文献报道选择方中各药味所含化学成分单体,均配置为25μg·mL^(-1)的单体溶液,加入A549细胞中进行孵育,使用ELISA法测定IL-1β水平;根据测定结果,选择具有明显调控作用的成分(盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷和紫菀酮),进而配制不同浓度的上述化合物单体溶液(12.5、25、50μg·mL^(-1)),测定IL-1β水平考察量效关系;上述成分均通过测试后,将其选为指标成分,建立HPLC同时测定方法,测定10批风寒咳喘方的含量,根据结果最终确定活性成分。结果风寒咳喘方各药味所含化学成分中,盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷和紫菀酮具有明显的IL-1β调控活性,且在12.5~50μg·mL^(-1)浓度范围内,活性与剂量呈明显相关性;含量测定结果显示,除紫菀酮未检出外,其余三种成分的含量分别为1.030~1.499 mg·mL^(-1)、0.729~0.892 mg·mL^(-1)、0.728~1.032 mg·mL^(-1)。结论风寒咳喘方对IL-β水平有明显调控作用,产生该功效的主要化学成分为毛蕊异黄酮葡萄糖苷、苦杏仁苷和紫菀酮。OBJECTIVE Through selection and determination of IL-1β regulatory compounds in FengHan KeChuan Formula, to establish a quality evaluation method closer to its efficacy and related mechanism.METHODS Components in the formula were selected according to the literature, and monomer solution(at the concentration of 25 μg·mL^(-1)) was added to A549 cell for cultivation, and used ELISA for IL-1β level determination;Compounds with obvious regulatory activity(namely ephedrina hydrochloridum, pseudoephedrine hydrochloride, Amygdalin and Shionone) were selected for further test, in which those compounds of a series of concentrations(12.5,25,50 μg·mL^(-1)) were prepared, and corresponding IL-1β levels were obtained to investigate their dose-effect relationship;by passing the tests, the above mentioned compounds were chosen as indexes for content determination using HPLC,and 10 batches of granule were analyzed to give final conclusion of the active component list.RESULTS Among all components in the ingredients that FengHan KeChuan Formula composed of, ephedrina hydrochloridum, pseudoephedrine hydrochloride, Amygdalin and Shionone had obvious IL-1β regulatory effects, and except Shionone, all the other three were in the concentration range of 12.5-50 μg·mL^(-1),their activities were dose-dependent.Content determination result showed that the content of the three components were 1.030-1.499 mg·mL^(-1),0.729-0.892 mg·mL^(-1) and 0.728-1.032 mg·mL^(-1),respectively.CONCLUSION FengHan KeChuan Formula had obvious IL-1β regulatory effect, and the main active components for such effect were Calycosin-7-glucoside, Amygdalin and Shionone.
