摘  要:目的建立能同时测定芍药苷以及芍药苷亚硫酸酯成分含量的HPLC法,为白芍中的芍药苷以及芍药苷亚硫酸酯成分含量测定提供一种新的方法。方法以Inertsil(4.6 mm×250 mm,5μm)ODS-2色谱柱为固定相;以乙腈-0.1%磷酸水溶液(14∶86)为流动相;检测波长为235 nm;流速为1.0 mL·min^(-1);进样量为10μL;柱温35℃。结果芍药苷、芍药苷亚硫酸酯分别在(0.0282~1.128μg、0.0196~0.784μg)的范围内与其峰面积呈现良好的线性关系,平均加样回收率分别为96.64%、96.78%;RSD分别为2.64%、2.49%。结论所建立的HPLC法能同时测定白芍及其饮片中的芍药苷以及芍药苷亚硫酸酯成分的含量,方法简单、准确,可考虑作为白芍药材及其饮片的质量控制方法。OBJECTIVE To establish an HPLC method that can simultaneously determine the content of paeoniflorin and paeoniflorin sulfite, and provide a new method for the determination of paeoniflorin and paeoniflorin sulfite in Paeoniae Radix Alba.METHODS Using Inertsil?(4.6 mm×250 mm, 5 μm) ODS-2 column was the stationary phase;using acetonitrile-0.1% phosphoric acid aqueous solution(14∶86) as the mobile phase;detection wavelength was 235 nm;flow rate was 1.0 mL·min^(-1);the injection volume was 10 μL and column temperature was 35 ℃.RESULTS The content of paeoniflorin and paeoniflorin sulfite showed a good linear relationship with its peak area in the range of(0.0282-1.128 μg, 0.0196-0.784 μg),respectively.The average sample recovery rates were 96.64% and 96.78%;RSD were 2.64% and 2.49%,respectively.CONCLUSION The established HPLC method can simultaneously determine the content of paeoniflorin and paeoniflorin sulfite in Paeoniae Radix Alba and its decoctions.The method was simple, accurate, it can be considered as the quality control method of Paeonia lactiflora and its decoction pieces.
