摘  要:目的观察石菖蒲核心代谢效应物α-细辛醇对癫痫幼鼠海马区神经元形态及星形胶质细胞活化的影响。方法将30只SD大鼠幼鼠随机分为正常组、模型组、溶剂对照组、阳性对照组和α-细辛醇组,每组6只。除正常组外,其余各组使用氯化锂—匹鲁卡品建立癫痫模型,溶剂对照组、阳性对照组和α-细辛醇组分别腹腔注射Tween80、司替戊醇和α-细辛醇50 mg/kg,正常组腹腔注射等体积生理盐水,每日给药3次,连续7 d。获取海马组织切片与组织块,尼氏染色观察幼鼠海马区神经元形态,免疫荧光染色及Westernblotting法检测各组幼鼠海马区星形胶质细胞活化标志物胶质纤维酸性蛋白(GFAP)。结果与正常组比较,模型组幼鼠海马CA1、CA3区锥体细胞层细胞间隙增大,神经元排列疏松紊乱,尼氏染色减弱;与模型组比较,α-细辛醇组和阳性对照组幼鼠海马CA1、CA3区锥体细胞边缘清晰,神经元排列整齐,尼氏染色加深。海马区GFAP蛋白平均光密度及相对表达量模型组、溶剂对照组>阳性对照组、α-细辛醇组>正常组(P均<0.05),模型组与溶剂对照组、阳性对照组与α-细辛醇组差异无统计学意义。结论α-细辛醇可减弱癫痫幼鼠海马区神经元细胞损伤,抑制星形胶质细胞活化。Objective To observe the effects ofα-asaronol,a core metabolic effector of acorus calamus,on neuronal morphology and astrocyte activation in hippocampus of epileptic young mice.Methods Thirty SD young rats were randomly divided into the normal group,model group,solvent control group,positive control group,andα-asaronol group,with 6 rats in each group.Except for the normal group,we used lithium-pilocarpine to establish epilepsy models in the other groups,and rats in the solvent control group,positive control group andα-asaronol group were injected intraperitoneally with Tween80,Stiripentol,andα-asaronol 50 mg/kg,respectively,and the rats in the normal group were intraperitoneally injected with the same volume of normal saline,three times a day,for 7 consecutive days.The sections and blocks were obtained,and the neuronal morphology of the hippocampus was observed by Nissl staining;immunofluorescence staining and Western blotting were used to detect the glial fibrillary acidic protein(GFAP),a marker of astrocyte activation,in hippocampus of young rats in each group.Results Compared with the normal group,the gap of pyramidal cell layer in hippocampal CA1 and CA3 regions increased in the model group,the arrangement of neurons was loose and disordered,and the Nissl staining was weak;compared with the model group,the pyramidal cell margins of theα-asaritol group and positive control group in hippocampal CA1 and CA3 regions were clear,the neurons were arranged neatly,and the Nissl staining was deepened.The MOD value and relative expression of GFAP protein in hippocampus were in the following order:the model group and solution control group>positive control group andα-asaronol group>normal group(all P<0.05).There were no statistical significant differences between the model group and solvent control group,or between the positive control group andα-asaronol group.Conclusion Theα-asaronol could attenuate neuronal cell damage in hippocampus of epileptic young rats and inhibit the activation of astrocytes.
