摘　　要：目的分析拓扑异构酶ⅡA(TOP2A)在宫颈鳞癌中的表达及其与患者临床病理特征和预后的关系。方法收集2014年2月至2016年9月在本院进行宫颈活检或手术中取得的宫颈鳞癌组织标本74例,选取同期在本院进行宫颈活检并经病理证实的正常宫颈组织标本53例为对照组。实时荧光定量PCR(qRT-PCR)法检测宫颈组织中TOP2A mRNA的表达水平;免疫组化法检测宫颈组织中TOP2A蛋白的表达情况;分析TOP2A蛋白阳性表达与宫颈鳞癌患者临床特征间的关系,并对患者出院后5年内进行随访,记录患者生存状态;Kaplan-Meier法生存曲线分析TOP2A表达与宫颈鳞癌患者预后的关系;COX回归分析影响宫颈鳞癌患者预后的因素。结果与对照组比较,宫颈鳞癌患者TOP2A mRNA表达水平及TOP2A蛋白阳性表达均显著增加(P<0.05);FIGO分期Ⅱ~Ⅲ期、深肌层浸润宫颈鳞癌组织TOP2A阳性表达率分别高于FIGO分期Ⅰ期、浅肌层浸润组织(P<0.05);宫颈鳞癌患者TOP2A阳性表达组5年内生存率(39.7%)低于阴性表达组(75.0%);多因素COX分析表明,肌层浸润深度、TOP2A阳性表达是影响宫颈鳞癌患者预后的危险因素(P<0.05)。结论TOP2A表达可能与宫颈鳞癌患者的预后有关,可能是预测宫颈鳞癌预后的潜在分子标志物。Objective:To analyze the expression of topoisomerase IIA(TOP2A)in cervical squamous cell carcinoma and its relationship with clinicopathological features and prognosis.Methods:Seventy-four samples of cervical squamous cell carcinoma tissue were collected from cervical biopsy or operation in our hospital from February 2014 to September 2016.During the same period,53 samples of normal cervical tissue confirmed by pathology in our hospital were collected as control group.The expression level of TOP2A mRNA was detected by real-time fluorescence quantitative PCR(qRT-PCR).The expression of TOP2A protein in cervical tissue was detected by immunohistochemistry.The relationship between TOP2A protein and clinical features of patients with cervical squamous cell carcinoma was analyzed.The patients were followed-up within 5 years after discharge,and the survival status of patients was recorded.Kaplan-Meier survival curve was used to analyze the relationship between the expression of TOP2A and the prognosis of patients with cervical squamous cell carcinoma.COX regression analysis was used to analyze the prognostic factors of patients with cervical squamous cell carcinoma.Results:The expression levels of TOP2A mRNA and the positive expression of TOP2A protein were significantly higher than those in the control group(P<0.05).The positive expression rate of TOP2A in FIGO stageⅡ-Ⅲand deep myometrial invasion cervical squamous cell carcinoma tissue was significantly higher than that in FIGO stage I and superficial myometrial invasion tissue(P<0.05).The 5-year survival rate of patients with cervical squamous cell carcinoma in positive TOP2A expression group(39.7%)was lower than that in negative expression group(75.0%).The multivariate COX analysis showed that the depth of myometrial invasion and the positive expression of TOP2A were the risk factors for the prognosis of patients with cervical squamous cell carcinoma(P<0.05).Conclusions:TOP2A expression may be related to the prognosis of patients with cervical squamous cell carc
