摘 要:目的探究五氯硝基苯(PCNB)亚急性暴露对SD大鼠卵巢甾体激素合成的影响。方法将30只3周龄雌性SD大鼠随机分为3组:给予50 mg·kg^(-1)·d^(-1)PCNB灌胃给药的为低剂量组,给予100 mg·kg^(-1)·d^(-1)PCNB灌胃给药的为高剂量组,给与等体积玉米油灌胃给药的为对照组,每组10只,连续灌胃28 d。用电化学发光法检测各组大鼠血清孕酮和E2水平;用蛋白质免疫印迹法(Western Blot)检测各组大鼠卵巢组织中3种甾体激素合成关键酶:类固醇激素合成急性调节蛋白(StAR)、胆固醇侧链裂解酶(P450scc)及芳香化酶(Aromatase)的表达情况。结果PCNB亚急性暴露期间,两实验组大鼠的体重变化、卵巢系数与对照组比较均无显著差异(P>0.05);两实验组大鼠血清孕酮水平均显著高于对照组,血清E2水平均显著低于对照组(P<0.05)。Western Blot结果显示,大鼠卵巢组织中StAR的表达量低剂量组>高剂量组>对照组,各组组间无统计学差异(P>0.05);大鼠卵巢组织中P450scc表达量高剂量组>对照组>低剂量组,各组组间无显著性差异(P>0.05);低、高剂量组大鼠卵巢组织中芳香化酶表达水平均显著低于对照组(P<0.05),并呈剂量依赖性下降。结论PCNB亚急性暴露可导致大鼠血清孕酮和E2水平发生改变,其机制可能与卵巢组织中甾体激素合成关键酶的表达量改变有关。Objective:To investigate the effect of subacute exposure to pentachloronitrobenzene(PCNB)on ovarian steroidogenesis in rats.Methods:Thirty 3-week-old female SD rats were randomly divided into three groups:the rats were given 50 mg·kg^(-1)·day^(-1) PCNB by gavage in low-dose group,the rats were given 100 mg·kg^(-1)·day^(-1) PCNB by gavage in high-dose group,and the rats were given the same volume of corn oil by gavage in control group,with 10 rats in each group for 28 consecutive days.The serum levels of progesterone and E2 of female rats after PCNB exposure were detected by electrochemiluminescence.The protein expression of steroidogenic acute regulatory protein(StAR),cytochrome P450scc enzyme(P450scc)and aromatase in ovarian tissue were detected by Western blot.Results:During subacute exposure to PCNB,there was no significant difference in body weight and ovarian coefficient between the two experiment groups and the control group(P>0.05).The serum levels of progesterone in two experiment groups were significantly higher than those in the control group(P<0.05),while E2 levels were significantly lower than those in the control group(P<0.05).The results of Western blot showed that the order of expression level of StAR in rat ovary was low-dose group>high-dose group>control group,and there was no significant difference among the groups(P>0.05).The order of expression level of P450scc was high-dose group>control group>low-dose group,and there was no significant difference among the groups(P>0.05).The expression levels of aromatase in ovaries of low-dose group and high-dose group were significantly lower than that of control group(P<0.05),and decreased in a dose-dependent manner.Conclusions:Subacute exposure to PCNB can lead to the changes of serum levels of progesterone and E2 in rats,which may be related to the changes in the expression of key enzymes of steroid hormone synthesis in ovarian tissue.
