摘 要:旨在探讨钩吻素子和钩吻素己对东莨菪碱诱导的轻度认知功能障碍小鼠模型的神经保护作用。将C57BL/6小鼠随机分为4组,即对照组、钩吻素子剂量组、钩吻素己剂量组、东莨菪碱模型组,每组6只。对照组和模型组给予生理盐水,其余组连续腹腔给药12 d,第8天开始除对照组外均注射东莨菪碱溶液,通过水迷宫试验观察小鼠在钩吻生物碱干预作用下的动物行为学指标变化。再通过HE染色观察钩吻素己对海马组织CA1、CA3、DG和PC区的病理学变化。水迷宫定位航行试验结果表明,模型组与对照组相比逃避潜伏期延长,钩吻素子组与模型组相比逃避潜伏期缩短,且都具有显著性差异;空间探索结果表明,模型组小鼠平台穿越次数和所在象限运动路程显著少于对照组,而钩吻素子组平台所在象限运动路程显著少于模型组。海马HE染色结果显示,钩吻素己组与对照组相比,海马CA1和CA3区细胞固化萎缩,整个细胞深染;CA3区和PC区的活细胞数量变少,但细胞坏死的数量增多。试验结果说明钩吻素子能改善东莨菪碱诱导的轻度认知功能障碍,改善小鼠学习记忆能力,而钩吻素己对东莨菪碱造成的认知功能障碍不具有保护作用,并对海马组织造成损伤。The aim of this study was to investigate the neuroprotective effects of koumine and gelsenicine on scopolamine induced mild cognitive impairment in mice.All mice were randomized into four groups(n=6/group):(1)control,(2)koumine,(3)gelsenicine and(4)scopolamine model.The control group and the model group were given normal saline,and the other groups were injected intraperitoneally for 12 days.Scopolamine solution was injected except the control group on the 8th day.The changes of animal behavior indexes of mice under the intervention of gelsemium alkaloids were observed by water maze test.The hippocampal CA1,CA3,DG and PC regions were observed by HE staining,under the intervention of gelsenicine.The results of water maze navigation experiment showed that the escape latency of the model group was longer than that of the control group,and the escape latency of the koumine group was shorter than that of the model group,and there were significant differences.The results of spatial exploration showed that the number of crossing the platform and the proporation distance in the quadrant in the model group were significantly less than those in the control group,while the proporation distance in the platform quadrant in the koumine group was significantly less than that in the model group.The results of hippocampal HE staining showed that compared with the control group,the cells in the CA1 and CA3 regions of the hippocampus were solidified and atrophied,and the whole cells were deeply stained.The number of cells in CA3 and PC regions decreased,but the number of necrotic cells increased.The results showed that koumine could improve the mild cognitive impairment induced by scopolamine and improve the ability of learning and memory in mice,while gelsenicine had no protective effect on the cognitive impairment caused by scopolamine and caused damage to the hippocampus tissue.
