摘 要:【目的】探讨雪胆多酚与氨基酸的联合抗氧化作用及对α-葡萄糖苷酶的抑制作用。【方法】采用Folin-Ciocalte法测定多酚含量。利用DPPH自由基、ABTS自由基、羟基自由基、超氧阴离子清除活性和铁还原能力法,评价雪胆多酚与组氨酸、缬氨酸、甘氨酸之间的联合抗氧化活性。并通过体外法测定雪胆多酚对α-葡萄糖苷酶的抑制作用。【结果】雪胆多酚、组氨酸、缬氨酸、甘氨酸、组氨酸与雪胆多酚联合、缬氨酸与雪胆多酚联合、甘氨酸与雪胆多酚联合均具有清除DPPH自由基、ABTS自由基、羟基自由基、超氧阴离子清除活性的能力,并具有一定的还原能力。抑制性实验结果表明,雪胆多酚对α-葡萄糖苷酶有抑制作用,属于可逆性抑制中的反竞争性抑制类型。【结论】利用体外抗氧化评价方法,明确了雪胆多酚与氨基酸之间具有协同抗氧化能力,以及雪胆多酚对α-葡萄糖苷酶具有抑制作用,为雪胆多酚的进一步开发利用提供理论依据。【Objective】The present research aimed to investigate the synergistic antioxidant of polyphenol from Hemsleya chinensis with amino acids,and the inhibition to α-glucosidase.【Method】The polyphenol content from H.chinensis was determined by Folin-Ciocalte method.The antioxidation activity of polyphenol from H.chinensis,histidine,valine and glycine were determined by DPPH,ABTS,hydroxyl radical,superoxide anion scavenging activity and reducing power,respectively.The inhibition effect of polyphenol from H.chinensis on α-glucosidase was determined in vitro.【Result】The results indicated that the synergistic antioxidant of polyphenol from H.chinensis,histidine,valine,glycine,histidine,and polyphenol from H.chinensis,valine and polyphenol from H.chinensis,glycine and polyphenol from H.chinensis had DPPH radical-scavenging activity,ABTS radical-scavenging activity,hydroxyl radicals-scavenging activity,superoxide anion radical-scavenging activity,and similar results had been shown in reducing power assay.【Conclusion】The antioxidant evaluation method in vitro was used to clarify the synergistic antioxidant capacity of polyphenol from H.chinensis and amino acids,and the inhibition effect of polyphenol from H.chinensis on α-glucosidase was demonstrated,which provided a theory for future development and utilization in H.chinensis.
