摘 要:目的:探讨有氧运动、抗阻运动、全身振动和骨骼肌电刺激对小鼠骨骼肌中应激诱导蛋白Sestrin2(SESN2)表达的影响以及运动激活SESN2/AMPK/PGC-1α通路改善心梗小鼠骨骼肌质量减少潜在机制。方法:10月龄清洁级雄性C57BL/6J野生型小鼠,随机分为安静对照组(CON)、有氧运动组(AE)、抗阻运动组(RE)、全身振动组(WBV)和骨骼肌电刺激组(ES),每组12只;假手术组(S)、心梗组(MI)、心梗有氧运动组(MAE)和心梗抗阻运动组(MRE),每组8只。运动结束后检测骨骼肌功能和质量,测试后取材,分离腓肠肌;Laminin染色测定骨骼肌横截面积(CSA);试剂盒检测丙二醛(malondialdehyde,MDA);Western Blot检测骨骼肌SESN2、p-AMPK、AMPK、PGC-1α、SOD1、SOD2、NF-κB、TNF-α、IL-1β、IL-10、Caspase-3、Bax、Bcl-2。结果:1)与CON组比较,AE组、RE组、WBV组和ES组骨骼肌相对质量和最大承载力显著增加,RE组和WBV组骨骼肌CSA显著增加,其中RE组干预效果最优。2)与CON组比较,AE组、RE组、WBV组和ES组骨骼肌SESN2、p-AMPK/AMPK、PGC-1α表达显著升高,其中RE组干预效果最优。3)与S组比较,MI组骨骼肌相对质量、最大承载力和骨骼肌CSA显著减少;SESN2、p-AMPK/AMPK、PGC-1α水平代偿性升高;MDA、NF-κB、TNF-α、IL-1β、Bax/Bcl-2、Caspase-3水平显著升高;SOD1、SOD2、IL-10水平显著降低。4)与MI组比较,MAE和MRE组骨骼肌相对质量、最大承载力、骨骼肌CSA显著增加;MRE组SESN2、p-AMPK/AMPK、PGC-1α表达进一步升高;MAE和MRE组MDA、NF-κB、TNF-α、IL-1β、Bax/Bcl-2、Caspase-3水平显著降低;SOD1、SOD2水平显著升高;MAE组IL-10水平显著升高。结论:有氧运动、抗阻运动、全身振动和骨骼肌电刺激可显著激活小鼠骨骼肌SESN2/AMPK/PGC-1α信号通路,提高小鼠骨骼肌质量,其中抗阻运动干预效果最优。有氧和抗阻运动可抑制心梗小鼠骨骼肌细胞氧化应激、炎症及细胞凋亡,抑制心梗小鼠骨骼肌质量减少,其�This study aims to investigate the effects of aerobic exercise,resistance exercise,whole-body vibration and muscle electrical stimulation on the expression of SESN2 in skeletal muscle and the potential mechanism for exercise to improve the mass of skeletal muscle in mice with myocardial infarction by activating SESN2/AMPK/PGC-1αsignaling pathway.Materials and methods:10-month-old male C57 BL/6 J wild mice are randomly divided into quiet control group(CON),aerobic exercise group(AE),resistance exercise group(RE),whole-body vibrationgroup(WBV)and skeletal muscle electrical stimulation group(ES),with 12 mice in each group;also,there are sham-operated group(S),myocardial infarction group(MI),MI with aerobic exercise group(MAE)and MI withresistance exercise group(MRE),with 8 mice in each group.After exercise,the function and mass of skeletal muscle are tested and the gastrocnemius muscle is separated.Laminin staining is performed to determine the crosssectional area of skeletal muscle(CSA).MDA is detected by assay kits.The protein expression of SESN2,p-AMPK,AMPK,PGC-1α,SOD1,SOD2,NF-κB,TNF-α,IL-1Β,IL-10,Caspase-3,Bax and Bcl-2 are detected by Western Blotting.Results:(1)Compared with the CON group,the relative mass and maximal carrying capacity of skeletal muscle significantly increase in the AE,RE,WBV and ES groups,especially the RE group.(2)Compared with the CON group,the protein expression of SESN2 and PGC-1αand the ratio of p-AMPK/AMPK signifi-cantly increase in the AE,RE,WBV and ES groups,and the cross-sectional area of the gastrocnemius muscle from the RE and WBV groups significantly increases,especially the RE group.(3)Compared with the S group,the relative mass,maximal carrying capacity and the cross-sectional area of the skeletal muscle in the MI group significantly decrease;the levels of SESN2,p-AMPK/AMPK and PGC-1αincrease in the MI group;the levels of MDA,NF-κB,TNF-α,IL-1β,Bax/Bcl-2 and Caspase-3 significantly increase in the MI group;the levels of SOD1,SOD2 and IL-10 significantly decrease in the
