摘  要:目的评价围术期穴位电刺激对脊柱手术大鼠树突状细胞(cDC)成熟的影响。方法SPF级健康雄性SD大鼠40只,7~8周龄,体重300~350 g,采用随机数字表法分为5组(n=8):对照组(C组)、手术组(S组)、疏密波组(D组)、连续波组(W组)和非经非穴组(N组)。D组和W组麻醉后立即电刺激双侧足三里和三阴交穴10 min,随后行脊柱手术并维持电针刺激至手术结束,手术时间约20 min。术后24和48 h电刺激双侧内关穴和太冲穴30 min。D组使用2/15 Hz疏密波,W组使用15 Hz连续波,N组使用2/15 Hz疏密波,但刺激部位为双侧髂嵴最高点连线上方15 mm,后正中线左右各旁开20 mm处。于术后48 h电刺激大鼠后处死,取脾并制备脾脏组织细胞悬液,采用流式细胞仪检测CD103+、CD11c+、MHC-Ⅱ+、CD80+和CD86+cDC百分比。结果与C组比较,D组和W组各指标差异均无统计学意义(P>0.05),S组MHC-Ⅱ+和CD86+cDC百分比降低,CD103+和CD80+cDC百分比升高,N组MHC-Ⅱ+、CD86+、CD11c+和CD103+cDC百分比降低(P<0.05);与S组比较,D组MHC-Ⅱ+、CD86+和CD103+cDC百分比升高,CD80+cDC百分比降低,W组MHC-Ⅱ+cDC百分比升高,CD80+cDC百分比降低,N组CD86+和CD11c+cDC百分比降低(P<0.05);与D组比较,W组各指标差异无统计学意义(P>0.05),N组CD86+和CD11c+cDC百分比降低(P<0.05)。结论围术期穴位电刺激可减轻脊柱手术对大鼠cDC成熟的抑制作用。Objective To evaluate the effect of perioperative electroacupuncture at acupoints on the maturation of dendritic cells in rats undergoing spinal surgery.Methods Forty SPF healthy male Sprague-Dawley rats,aged 7-8 weeks,weighing 300-350 g,were divided into 5 groups(n=8 each)using a random number table method:control group(group C),surgery group(group S),disperse-dense wave group(group D),continuous wave group(group W),and non-acupoint group(group N).After anesthesia in D and W groups,electro-stimulation of bilateral"Zusanli"(ST36)was performed for 10 min after anesthesia,followed by spinal surgery,the electro-stimulation was maintained until the end of operation,and the operation time was about 20 min.Electro-stimulation of bilateral"Neiguan"and"Taichong"acupoints was performed for 10 min starting from 24 and 48 h after surgery.Disperse-dense wave 2/15 Hz was used in group D,continuous wave 15 Hz in group W,and disperse-dense wave 2/15 Hz in group N,but the stimulation site was the point 15 mm above the line of the highest point of the bilateral iliac crest,and the point 20 mm lateral to the left and right sides of the posterior median line.The rats were sacrificed after electro-stimulation at 48 h after operation,and the spleen was obtained to prepare the tissue cell suspension.The percentage of CD103+,CD11c+,MHC-Ⅱ+,CD80+and CD86+cDC was detected by flow cytometry.Results Compared with group C,no significant change was found in each parameter in D and W groups(P>0.05),the percentage of MHC-Ⅱ+,CD86+and CD103+cDC was significantly decreased,and the percentage of CD80+cDC was increased in group S,and the percentage of MHC-Ⅱ+,CD86+,CD11c+and CD103+cDC was significantly decreased in group N(P<0.05).Compared with group S,the percentage of MHC-Ⅱ+,CD86+and CD103+cDC was significantly increased,and the percentage of CD80+cDC was decreased in group D,the percentage of MHC-Ⅱ+cDC was significantly increased,and the percentage of CD80+cDC was decreased in group W,and the percentage of CD86+and CD11c+cDC was signific
