摘  要:目的通过超声生物显微镜(ultrasound biomicroscopy,UBM)检查载脂蛋白E基因敲除(apolipoprotein E gene knock out,ApoE^(-/-))小鼠的左颈总动脉,并与病理学检测结果对比,研究UBM检查ApoE^(-/-)小鼠左颈总动脉的动脉粥样硬化(atherosclerosis,AS)特点和其有效的检测指标。方法采用自身前后对照设计,通过高脂喂养ApoE^(-/-)小鼠进行AS模型复制。分别在模型复制的第16周、第20周和第24周,采用UBM检查并比较小鼠左颈总动脉的内-中膜厚度(intima-media thickness,IMT)和管腔狭窄程度(stenosis index,SI)指标的变化,并与病理学检测结果比较。结果UBM检查显示ApoE^(-/-)小鼠左颈总动脉的IMT和SI指标随AS模型时间的延长而递增。在ApoE^(-/-)小鼠高脂喂养第16周、第20周和第24周的IMT分别为(0.11±0.01)mm、(0.14±0.01)mm和(0.15±0.01)mm,SI分别为(61±11)%、(73±8)%和(77±4)%,与第16周比较,第20周和第24周的IMT以及SI均显著增高(P<0.05)。第24周的病理学检测的IMT结果与UBM检测结果比较差异无统计学意义[(0.18±0.04)mm vs(0.15±0.01)mm,P>0.05],且与UBM检查结果呈正相关(r=0.80,P=0.016)。结论UBM成像作为一种无创检测方法可准确评价ApoE^(-/-)小鼠颈总动脉的动脉粥样硬化的病变程度。Objective To detect developing characters and explore effective parameters of atherosclerosis(AS)at left common carotid artery(LCCA)in apolipoprotein E gene knockout(ApoE^(-/-))mice.Methods A self-control study was conducted.The AS model was established by feeding ApoE^(-/-)mice with high-fat diet.The ultrasound biomicroscope(UBM)method was applied to detect the intima-media thickness(IMT)and stenosis index(SI)of the LCCA in ApoE^(-/-)mice at the 16^(th),20^(th),and 24^(th) weeks respectively.And the histopathology method was applied to compare with the results detected by the UBM method at the 24th week.Results The UBM detection showed that the values of IMT and SI in LCCA were progressed as AS model developing with time course in ApoE^(-/-)mice.At the 16^(th),20^(th),and 24^(th) weeks,the IMTs were(0.11±0.01)mm,(0.14±0.01)mm,and(0.15±0.01)mm respectively,and the SIs were(61±11)%,(73±8)%,and(77±4)%respectively.Both the values of IMT and SI at the 20th and 24th week were significantly increased than those at the 16th week(all P<0.05).The value of IMT detected by the histopathology was consistent with that by the UBM method[(0.18±0.04)mm vs(0.15±0.01)mm,P>0.05)]and showed a positive correlation(r=0.80,P=0.016).Conclusion The UBM imaging technique could be a non-invasive observation method to evaluate the AS development of LCCA in ApoE^(-/-)mice accurately.
