摘 要:目的探究天香丹胶囊含药血浆对氧化损伤心肌细胞的影响。方法制备含药血浆和原代培养心肌细胞,用H_(2)O_(2)诱导心肌细胞建立氧化损伤模型,将细胞分为对照组、模型组(100μmol/L H_(2)O_(2))、空白血浆组预处理24 h、天香丹胶囊含药血浆低、中、高浓度组预处理24 h,用MTT法检测天香丹胶囊含药血浆对100μmol/L H_(2)O_(2)氧化损伤心肌细胞2 h的细胞活力;Hoechst 33342染色法观察心肌细胞凋亡形态变化;生化试剂法测定心肌细胞中总抗氧化能力(total antioxidant capacity,TAOC),超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)的含量;Western blot法检测心肌细胞中核因子E2相关因子2(nuclear factor E2-related factor 2,Nrf2)和三磷酸肌醇受体(Inositol 1,4,5-trisphosphate receptor,IP3R)的蛋白表达水平。结果与对照组比较,模型组心肌细胞的存活率、T-AOC和SOD水平显著降低并有明显凋亡形态变化,心肌细胞中MDA含量明显增加,差异有统计学意义(P<0.05);与模型组比较,天香丹胶囊含药血浆组预处理心肌细胞后,可明显增强心肌细胞活力,提高T-AOC和SOD水平,减轻细胞凋亡变化和降低MDA含量,差异有统计学意义(P<0.05)。Western blot结果显示,与对照组比较,模型组心肌细胞的Nrf2蛋白表达水平明显降低,而IP3R蛋白表达水平明显增加,差异有统计学意义(P<0.05);与模型组比较,天香丹胶囊含药血浆组预处理心肌细胞后,可明显提高Nrf2的表达水平,降低IP3R的表达水平,差异有统计学意义(P<0.05)。结论天香丹胶囊含药血浆对氧化损伤的心肌细胞有保护作用。Objective To investigate the effect of Tianxiangdan capsule containing plasma on oxidative injury of cardiomyocytes.Methods To prepare drug-containing plasma and primary cultured cardiomyocytes;The oxidative injury model was established by H_(2)O_(2)on myocardial cells,and the cells were divided into control group,model group(100μmol/L H_(2)O_(2)),blank plasma group,and low,medium and high concentration groups of Tianxiangdan capsule containing plasma.All groups were pretreated for 24 h,and MTT assay was used to detect the cell viability of Tianxiangdan capsule containing plasma against 100μmol/L H_(2)O_(2)damaged myocardial cells for 2 h.The morphological changes of apoptosis were observed by Hoechst 33342 staining.The contents of total antioxidant capacity(T-AOC),superoxide dismutase(SOD)and malondialdehyde(MDA)in myocardial cells were determined by biochemical reagent method.The protein expression levels of nuclear factor E2-related factor 2(Nrf2)and Inositol 1,4,5-trisphosphate receptor(IP3R)in the cells were detected by Western blotting.Results Compared with the control group,the cell survival rate,T-AOC and SOD levels in the model group were significantly decreased,and the apoptotic morphology was significantly changed,while the content of MDA in the model group was significantly increased,with statistical significance(all P<0.05);compared with the model group,cell viability was enhanced for pretreatment of cardiomyocytes in Tianxiangdan capsule containing plasma group significantly,T-AOC and SOD levels were increased,cell apoptosis was reduced and MDA content was decreased,with statistical significance(all P<0.05).Western blotting results showed that compared with the control group,the expression level of Nrf2 protein in the model group was significantly decreased,while the expression level of IP3R protein was significantly increased,with statistical significance(both P<0.05);compared with the model group,the expression level of Nrf2 was increased and the expression level of IP3R were significantly decre
