摘 要:目的:探讨小檗碱对高脂加链脲佐菌素(streptozotocin,STZ)诱导的糖尿病大鼠的影响。方法:70只SD大鼠中随机选取10只作为空白对照组,其余接受高脂饮食加STZ诱导并分为糖尿病模型组(B组)、阳性对照组(C组)、小檗碱低剂量组(D组)、小檗碱中剂量组(E组)、小檗碱高剂量组(F组)。C组给予盐酸二甲双胍(0.250 mg·g^(-1)),D、E、F组给予小檗碱(0.125 mg·g^(-1)、0.250 mg·g^(-1)、0.375 mg·g^(-1)),A、B组给予等量生理盐水灌胃。结果:与B组比较,治疗后C、D、E、F组空腹血糖(fasting blood glucose,FBG)、胰岛素抵抗指数(homeostasis model-insulin resistance,HOMA-IR)、血清总胆固醇(total cholesterol,TC)、三酰甘油(triglycerides,TG)、瘦素(leptin,LEP)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)及TNF-α、IL-6的mRNA表达水平均显著降低(P<0.05),空腹胰岛素(fasting insulin,FINS)、胰岛素敏感指数(homeostasis model-insulin sensitivity,HOMA-IS)、血清脂联素(adiponectin,ADPN)均显著提高(P<0.05),高剂量小檗碱治疗效果优于低、中剂量,与二甲双胍效果相当。结论:小檗碱可降低高脂饮食加STZ诱导的糖尿病大鼠的血糖,调控脂代谢与胰岛素水平。Objective:To investigate the effect of berberine on diabetic rats induced by high-fat and streptozotocin(STZ).Methods:Ten of the 70 SD rats were randomly selected as the blank control group,and the rest received high-fat diet plus STZ induction and were divided into diabetes model group(group B),positive control group(group C),low-dose berberine group(group D),middle-dose berberine group(group E)and high-dose berberine group(group F).Group C was given metformin hydrochloride(0.250 mg·g^(-1)),groups D,E,and F were given berberine(0.125 mg·g^(-1),0.25 mg·g^(-1),0.375 mg·g^(-1));groups A and B were given the same amount of normal saline intragastrically.Results:Compared with group B,the levels of fasting blood glucose(FBG),homeostasis model-insulin resistance(HOMA-IR),total cholesterol(TC),triglycerides(TG),leptin(LEP),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)as well as TNF-αmRNA and IL-6 mRNA in groups C,D,E and F were significantly reduced(P<0.05),the levels of fasting pancreas Insulin(FINS),homeostasis model-insulin sensitivity(HOMA-IS)and adiponectin(ADPN)were significantly increased(P<0.05),and the therapeutic effect of high-dose berberine was better than that of low-dose and medium-dose berberine,which is equivalent to the effect of metformin.Conclusion:Berberine is able to reduce blood sugar,regulate lipid metabolism and insulin levels in diabetic rats induced by high-fat diet plus STZ.
