摘  要:目的:研究昆布多糖(laminarin,LA)对柯萨奇病毒B3(coxsackievirus group B type3,CVB3)诱导的新生小鼠病毒性心肌炎的调节作用。方法:将100只BALB/c新生小鼠随机分为对照组(n=20)、LA组(n=20)、CVB3组(n=30)和CVB3+LA组(n=30)。CVB3组和CVB3+LA组腹腔注射CVB3进行模型复制,造模后LA组和CVB3+LA组腹腔注射LA(50 mg·kg^(-1))干预,连续注射12 d。检测新生小鼠心脏功能,血清诱导型一氧化氮合酶(iNOS)和白细胞介素-10(interleukin-10,IL-10)、肌红蛋白(myohemoglobin,Mb)、肌酸激酶同工酶-MB(creatine kinase-MB,CK-MB)和心肌肌钙蛋白I(cardiac troponin I,cTn I)水平;HE染色观察心肌组织病理变化;TUNEL法检测心肌细胞凋亡;Western Blot法检测心肌组织IL-6、Bax、Bcl-2、NF-κB p65、p-p65、TNF-α表达;RT-PCR法检测心肌组织CVB3 mRNA和IFN-γmRNA的表达。结果:与CVB3组相比,CVB3+LA组新生小鼠尾部动脉收缩压(systolic blood pressure,SBP)、心率(heart rate,HR)和左心室收缩压(left ventricular systolic pressure,LVSP)指数明显升高(P<0.01),Mb、CK-MB、cTn l、iNOS水平明显下降(P<0.01),心肌细胞凋亡率显著下降(P<0.01),IL-10、Bcl-2和IFN-γmRNA的表达显著升高(P<0.01),CVB3 mRNA的水平及p-p65、TNF-α、IL-6和Bax蛋白表达显著下降(P<0.01),同时可改善心肌组织损伤。结论:LA可能通过抑制炎症反应和细胞凋亡保护CVB3诱导病毒性心肌炎新生小鼠的心脏功能。Objective:To explore the regulation effects of laminarin(LA)in inhibiting coxsackievirus group B type3(CVB3)induced viral myocarditis in neonatal mice.Methods:A total of 100 BALB/c neonatal mice were randomly divided into control group(n=20),LA group(n=20),CVB3 group(n=30)and CVB3+LA group(n=30).CVB3 group and CVB3+LA group were intraperitoneally injected to copy the model.After molding,the LA group and CVB3+LA group were intraperitoneally injected with 50 mg·kg^(-1) laminarin for 12 d.Cardiac function,levels of iNOS and IL-10,Mb,CK-MB,and cTn I in serum,and protein expression levels of Bax/Bcl-2,NF-KB p65,p-p65,and TNF-αin myocardial tissues were measured in each group.HE staining and TUNEL staining were used to analyze the pathological features of myocardial tissues.Results:Compared with CVB3 group,the MAP,HR,and LVSP indexes were significantly increased in CVB3+LA group(P<0.01),and the expression levels of serum Mb,CK-MB,cTn l,iNOS were significantly decreased(P<0.01),and the apoptosis rate of cardiomyocytes was significantly decreased(P<0.01),the expression of IL-10,Bcl-2 and IFN-γmRNA was significantly increased(P<0.01),while the CVB3 mRNA,p-p65,TNF-α,IL-6 and Bax protein were significantly down-regulated(P<0.01),meantime,it improves the pathological damage of myocardial tissue.Conclusion:LA may improve the cardiac function of CVB3-induced viral myocarditis in neonatal mice by inhibiting inflammation and apoptosis.
