摘 要:目的:基于槲皮素与DES(4,4'-二叠氮二苯乙烯-2,2'-二磺酸二钠四水合物)原位反应的荧光特性,建立测定槐米中槲皮素含量的方法。方法:在激发波长(Ex)为383 nm,狭缝宽度2 nm的条件下测试不同浓度槲皮素对照品与DES混合溶液的荧光发射光谱,获得槲皮素对照品浓度与荧光强度之间的线性关系,测定槐米中槲皮素的含量。结果:在最佳测定条件下,槲皮素的浓度与溶液的荧光强度在0.25~4.00 mmol·L^(-1)范围内具有良好的线性关系,检测限为5.45×10-3 mmol·L^(-1),线性相关系数为0.9996。结论:基于还原态DES荧光特性建立的检测槐米中槲皮素的方法具有简便、快捷、灵敏等特点,可用于槐米中槲皮素含量的快速检测,为槐米药材的质量评价提供参考。Objective:Based on the fluorescence characteristics of the in-situ reaction between quercetin and DES(4,4'-diazide stilbene-2,2'-disodium disulfonate tetrahydrate),a method for the detection of Quercetin in sophora japonica was established.Methods:The fluorescence emission spectra of quercetin and DES mixed solutions with different concentrations were tested under the excitation wavelength(Ex)of 383 nm and the width of the slits of 2 nm.The linear relationship between the concentration of quercetin and fluorescence intensity was obtained and used to analyze the content of quercetin in Sophora japonica.Results:Under optimal conditions,the concentration of quercetin and the fluorescence emission intensity of the mixed solution showed a good linear relationship in the range of 0.25-4.00 mmol·L^(-1),and the detection limit was as low as 5.45×10-3 mmol·L^(-1)(R2=0.9996).Conclusion:The method for detecting Quercetin in sophora japonica based on the fluorescence characteristics of reduced DES is simple and sensitive.It can be used for the quick detection of Quercetin in sophora japonica,which provides a reference for the quality evaluation of sophora japonica.
