摘 要:目的:研究川芎嗪对体外培养大鼠成纤维细胞增殖及碱性成纤维细胞因子(Basic fibroblast growth factor,bFGF)、转化生长因子β_(1)(Transforming growth factorβ_(1),TGF-β_(1))表达的影响,从而探讨川芎嗪抑制瘢痕形成的作用机制。方法:体外分离培养大鼠皮肤成纤维细胞,采取MTT法及荧光染色检测不同浓度的川芎嗪对大鼠成纤维细胞增殖能力的影响,ELISA法测定大鼠皮肤瘢痕成纤维细胞TGF-β_(1)和bFGF的表达量。结果:在三种川芎嗪浓度条件下(1mg/ml,10mg/ml,50mg/ml),大鼠皮肤成纤维细胞增殖能力有不同程度的抑制作用,抑制作用与川芎嗪浓度成正比,在50mg/ml浓度时,抑制作用最为明显。采用ELISA法测定培养上清TGF-β_(1)和bFGF的分泌量,发现实验组两者含量均明显低于对照组,其中浓度为50mg/ml时,含量最低(P<0.05)。结论:川芎嗪能够明显抑制体外培养的大鼠皮肤成纤维细胞的增殖能力,降低TGF-β_(1)和bFGF的表达量,从而在一定程度上抑制大鼠皮肤成纤维细胞的活性。Objective To investigate the influence of ligustrazine on the proliferation and expresson of TGF-β_(1) and bFGF for human normal and hypertrophic scar dermal fibroblasts cultured in vitro and to explore the relevant mechanism.Methods The rat skin fibroblasts were isolated and cultured in vitro,and MMT assay was used to detect the effects of ligustrazine at various concentrations on the rat skin scar fibroblasts in vitro.ELISA was carried out to determine the contents of TGFβ_(1) and bFGF in rat skin scar fibroblasts.Results Under 3 ligustrazine concentrations(1 mg/ml,10 mg/ml,50 mg/ml),the rat skin fibroblasts were suppressed to various degrees,and such suppression was positively correlated with concentration.Typically,the most obvious suppression was attained at the concentration of 50 mg/ml.Besides,TGF-β_(1) and bFGF contents were measured by ELISA,the results suggested that,the expression contents in experiment group were lower than those in control group,and the lowest contents were detected at the concentration of 50 mg/ml(P<0.05).Conclusion Ligustrazine markedly suppresses the activity of rat skin fibroblasts cultured in vitro,and reduces the TGF-β_(1) and bFGF contents.Typically,the efficacy peaks under a relatively suitable concentration,thus partially suppressing the activity of rat fibroblasts.
