摘  要:目的基于lncRNA-mRNA网络探讨党参水提物对衰老小鼠胰腺的保护作用及分子机制。方法采用颈背部皮下注射D-半乳糖溶液制备衰老小鼠模型。将SPF级昆明种小鼠随机分为正常对照组、模型组和党参低、中、高剂量组,各给药组给予相应剂量药液灌胃,连续42 d。HE染色观察胰腺组织形态;筛选差异基因进行GO、KEGG通路分析;选取衰老后和党参干预后共同变化基因构建lncRNA-mRNA共表达网络;采用RT-qPCR验证网络中关键基因。结果模型组小鼠胰腺组织形态明显异常,党参低、中、高剂量组小鼠胰腺组织形态均有不同程度改善,党参高剂量组改善最明显;芯片聚类分析显示,衰老后和高剂量党参干预后lncRNAs和m RNAs发生明显变化;生物信息学分析显示,衰老后和党参干预后差异基因相同GO条目有脂肪酸代谢和线粒体,相同KEGG通路有代谢通路;对lncRNA-mRNA网络中关键基因Eci1、Dpep1、Serpini2进行RT-qPCR验证,其结果与基因芯片分析一致。结论 lncRNA-mRNA网络在党参保护衰老小鼠胰腺中发挥重要作用。Objective To explore the protective effect and molecular mechanism of Codonopsis Radix on the pancreas of D-galactose-induced aging mice based on the lncRNA-mRNA network. Methods The aging model was prepared by subcutaneous injection of D-galactose solution on the back of the neck. SPF Kunming mice were randomly divided into normal control group, model group and low-, medium-, and high-dosage Codonopsis Radix groups. Each administration group was given the corresponding dosage of liquid medicine by gavage for 42 consecutive days. HE staining was used to observe the morphology of pancreas;differential genes were screened for GO analysis and KEGG pathway analysis;genes that change together after aging and after the intervention of Codonopsis Radix were selected to construct lncRNA-mRNA co-expression network;RT-qPCR was used to verify key genes in the network. Results The pancreatic tissue morphology of mice in the model group was obviously abnormal. After the intervention of low-, medium-, and high-dosage Codonopsis Radix, the tissue structure of the pancreas in each group was improved to different degrees, and the effect of high-dosage Codonopsis Radix group was the most significant. Microarray cluster analysis showed that lncRNAs and mRNAs changed significantly after aging and the intervention of high-dosage Codonopsis Radix. Bioinformatics analysis of different genes after aging and the intervention of Codonopsis Radix, the same GO entries had fatty acid metabolism and mitochondria, and the same KEGG pathway had matabolic pathways. The key genes Eci1, Dpep1, and Serpini2 in the lncRNA-mRNA network were verified by RT-qPCR, and the results were consistent with the gene chip. Conclusion The lnc RNA-m RNA network plays an important role in the protection of Codonopsis Radix on pancreas of aging mice.
