补脑止痫散对戊四氮致痫大鼠行为学及海马组织IL-1β、IL-6的影响研究

摘　　要：目的:通过观察复方补脑止痫散对戊四氮癫痫模型大鼠行为学影响、海马组织炎性因子白介素表达的变化,探讨其防治癫痫疾病的作用机制。方法:随机选取实验大鼠作为补脑止痫散防治组,并给予其补脑止痫散灌胃干预;7 d后,随机选取实验大鼠作为空白组,除空白组外,将实验大鼠进行戊四氮癫痫模型点燃,除防治组外,将其余点燃成功的大鼠随机分为模型组、补脑止痫散组;记录灌胃治疗前、后大鼠一般状态及痫性发作情况,免疫组化法检测海马组织炎性因子白介素表达变化情况。结果:大鼠戊四氮癫痫模型点燃后,模型组、防治组及补脑止痫组较空白组大鼠体质量均明显减小、海马组织的IL-1β及IL-6阳性细胞数明显增高、平均灰度值明显降低,均有高度显著性差异(P<0.01);灌胃治疗后,与模型组比较,防治组、补脑止痫散组大鼠的体质量均明显增加、发作潜伏期均明显延长、发作级别显著降低,差异具有高度显著性差异(P<0.01),防治组与模型组比较IL-1β及IL-6阳性细胞数及平均灰度值均有高度显著性差异(P<0.01),补脑止痫散组与模型组比较IL-1β及IL-6阳性细胞数及平均灰度值均有显著性差异(P<0.05)。结论:补脑止痫散可明显改善戊四氮致痫大鼠一般状态、减轻大鼠痫性发作,具有降低致痫大鼠海马组织IL-1β、IL-6细胞因子表达的作用,抑制炎性反应,其治疗及预防癫痫疾病可能与此机制有关。Objective:To observe the effects of Bunao Zhixian Powder on behavior changes and the expressions of IL-1βand IL-6 in hippocampus of epileptic rats induced by Pentylenetetrazol,thus to investigate the mechanism of Bunao Zhixian powder in preventing and treating epilepsy.Methods:7 days before modeling,Bunao Zhixian Powder was given to the rats in the prevention group.The epilepsy model was ignited by Pentylenetetrazol.After successful modeling,the rats were randomly divided into the model group and the treatment group.The treatment group was treated with Bunao Zhixian Powder.The normal rats was taken as the blank control.General state and epileptic seizures were observed before and after the intervention in the groups.The expressions of IL-1βand IL-6 in hippocampus were detected by immunohistochemistry.Results:After the ignition of the epilepsy,the body weight was significantly decreased,the positive cells number of IL-1βand IL-6 in the hippocampus was significantly increased and the mean gray value was significantly decreased in the model group,the prevention group and the treatment group compared to those in the blank control group(P<0.01).After intragastric administration,the body weight of the rats were significantly increased,the onset latency was significantly prolonged,and the onset grade was significantly decreased in the prevention group and the treatment group compared to those in the model group(P<0.01).There were statistical differences in the positive cells number and the average gray value of IL-1βand IL-6 between the prevention group and the model group(P<0.01);the differences could also been seen between the treatment group and the model group(P<0.05).Conclusion:Bunao Zhixian Powder can obviously improve the general state of the epileptic rats induced by Pentylenetetrazol and alleviate epileptic seizures in the rats.It can reduce the cytokines expressions of IL-1βand IL-6 in the hippocampus of the epileptic rats and inhibit the inflammatory responses,which may be related to the mechanism of Bun
