摘 要:目的分析血浆中微小核糖核酸(microRNAs, miRNAs)与2型糖尿病(type 2 diabetes mellitus, T2DM)患病风险的相关性。方法 2018年7~12月,采用病例对照研究,纳入病例与对照各40例,开展问卷调查、体格测量及生化指标检测,运用实时荧光定量PCR(RT-qPCR)检测相关miRNAs,应用Logistic回归进行患病风险评估。结果 T2DM组miR-16、miR-17、miR-185及miR-328表达水平比对照组(NC)高,差异有统计学意义(P<0.05)。MiR-16及miR-188分别与HDL-C及TG呈负相关(P<0.05)。与NC组相比,miR-16、miR-17、miR-185及miR-328分别使T2DM组患病风险提高了0.70倍(OR=1.70,95%CI:1.01~2.84)、1.15倍(OR=2.15,95%CI:1.01~4.57)、1.57倍(OR=2.57,95%CI:1.25~5.27)及0.66倍(OR=1.66,95%CI:1.06~2.61),调整相关混杂后,患病风险进一步提高。结论血浆中高表达的miR-16、miR-17、miR-185及miR-328可能与T2DM患病风险相关,miRNAs是T2DM较好的生物标志物。Objective To analyze the correlation between miRNAs in plasma and risk of T2 DM. Methods From July to December, 2018,a case-control study was used, and 40 cases in the control group and 40 cases in the case control group were included.Questionnaire survey, physical measurement and biochemical indicators were conducted.Real-time fluorescent quantitative PCR(RT-qPCR) was used to detect the related miRNAs, and Logistic regression was used to assess the disease risk. Results The expression level of miR-16,miR-17,miR-185 and miR-328 were higher in plasma of T2 DM,and the difference were statistically significant(P<0.05).MiR-16 and miR-188 were negatively correlated with HDL-Cand TG(P<0.05),respectively.Compared with the NC group, the risk of T2 DM was increased 0.70 times(OR=1.70,95%CI:1.01-2.84),1.15 times(OR=2.15,95%CI:1.01-4.57),1.57 times(OR=2.57,95%CI:1.25-5.27) and 0.66 times(OR=1.66,95%CI:1.06-2.61),respectively, by miR-16,miR-17,miR-185 and miR-328,and the disease risk was further increased after adjusting the related confounding factors. Conclusion Higher expression of mir-16,miR-17,miR-185 and miR-328 in plasma may be associated with the risk of T2 DM.miRNAs are good biomarkers for T2 DM.
