摘  要:目的探讨核黄素转运体2(RFT2)在肺腺癌中的表达、预后意义及其生物学功能。方法检索GEPIA数据库中有关肺腺癌的数据,分析肺腺癌组织与癌旁组织中RFT2 mRNA表达的差异。采用Kaplan-Meier方法分析RFT2高、低表达组肺腺癌患者总生存时间(OS)、无病进展生存时间(DFS)的差异。利用The Human Protein Atlas分析肺腺癌组织和正常肺组织中RFT2蛋白表达的差异。通过String和WebGestalt数据库分析与RFT2表达相关的蛋白及其生物学功能。结果肺腺癌组织中RFT2 mRNA的表达水平明显高于正常肺组织,差异有统计学意义(P<0.01)。不同分期肺腺癌组织中RFT2 mRNA表达比较差异无统计学意义(P>0.05)。RFT2 mRNA高表达患者OS缩短(HR=1.4,P<0.05),DFS无明显改变(HR=1.0,P>0.05)。The Human Brotein Atlas数据库分析显示,肺腺癌组织RFT2蛋白呈高表达。String及WebGestalt分析发现,与RFT2基因相关的蛋白主要位于细胞膜中,参与细胞的生物调节、代谢和蛋白、离子的结合过程。结论RFT2在肺腺癌组织中呈现高表达,与肺腺癌预后差相关,有可能成为肺腺癌重要治疗靶点。Objective To explore the expression,prognostic significance and biological function of riboflavin transporter 2(RFT2)in lung adenocarcinoma.Methods Search the data about lung adenocarcinoma in the GEPIA database,and analyze the differences in the expression of RFT2 mRNA and protein in lung adenocarcinoma tissues and adjacent tissues.The Kaplan-Meier method was used to analyze the difference in overall survival and progression-free survival of lung adenocarcinoma patients with high and low RFT2 expression groups.Analyze RFT2 expression related proteins and biological functions through String and WebGestalt databases.The Human Protein Atlas was used to analyze the expression of RFT2 protein in lung adenocarcinoma tissue and normal lung tissue.Results The expression level of RFT2 mRNA in lung adenocarcinoma tissue was significantly higher than that in normal lung tissue,and the difference was statistically significant(P<0.01).There was no significant difference in the expression of RFT2 mRNA in lung adenocarcinoma tissues of different stages(P>0.05).The overall survival of patients with high expression of RFT2 mRNA was shortened(HR=1.4,P<0.05),and there was no significant change in disease-free survival(HR=1.0,P>0.05).Analysis of The Human Brotein Atlas database showed that RFT2 protein was highly expressed in lung adenocarcinoma tissue.String and WebGestalt analysis found that the proteins related to the RFT2 gene are mainly located in the cell membrane and participate in the cell's biological regulation,metabolism,and the binding process of proteins and ions.Conclusion RFT2 is highly expressed in lung adenocarcinoma tissues,which is related to the poor prognosis of lung adenocarcinoma,and may become an important therapeutic target for lung adenocarcinoma.
