摘 要:目的明确颐脑解郁方对血管性痴呆(vascular dementia,VD)的治疗作用,并探索颐脑解郁方改善VD认知障碍是否与调节甲状腺激素的变化相关。方法大鼠VD模型选择改良版的线栓法造成大鼠脑血管性病变后,不予任何干预4周后经跳台实验筛选出VD模型。实验分为正常组、假手术组、VD模型组、VD中药组4组,每组大鼠15只。VD中药组大鼠灌胃中药颐脑解郁方,正常组、假手术组、VD模型组大鼠灌胃0.9%生理盐水,连续灌胃4周。灌胃1周与4周末次行跳台实验及Morris水迷宫相关检测后,每组随机选择6只大鼠麻醉处死,取下丘脑HE染色观察其病理形态。灌胃4周末次行为学检测后,随机选择8只大鼠进行腹主动脉取血,采用放射免疫法检测血清促甲状腺素(thyroid-stimulating hormone,TSH)、游离三碘甲状腺原氨酸(free triiodothyronine,FT3)、游离甲状腺素(free thyroxine,FT4)的含量。结果跳台实验:与正常组比较,VD模型组及VD中药组大鼠灌胃1周和4周逃避潜伏期时间显著减少,错误次数显著升高(P<0.01);与VD模型组比较,VD中药组大鼠灌胃1周和4周逃避潜伏期时间明显增多,4周错误次数明显减少(P<0.05)。Morris水迷宫实验:与正常组比较,VD模型组及VD中药组大鼠灌胃4周逃避潜伏期时间明显增多(P<0.05,P<0.01),VD模型组大鼠灌胃1周穿越平台次数显著下降(P<0.01),VD模型组及VD中药组大鼠灌胃4周后穿越平台次数明显下降(P<0.05);与VD模型组比较,VD中药组大鼠灌胃1周和4周逃避潜伏期时间减少,穿越次数增多(P>0.05)。HE染色光镜下显示,VD中药组较VD模型组大鼠下丘脑细胞变性减轻,但仍见大片液化性坏死灶,呈筛网状结构,坏死区内神经元细胞变性、坏死,小胶质细胞增生,炎症细胞浸润,坏死边缘带窄,灌胃4周较1周大鼠下丘脑细胞变性改善明显。放射免疫法检测显示,与正常组比较,VD模型组、VD中药组FT3、FT4含量显著下降(P<0.01),Objective To further clarify the therapeutic effect of Yinao Jieyu formula on vascular dementia(VD).In this study,the relationship between VD and hypothalamic-pituitary-thyroid axis(HPT)function was studied through exploring hormone level changes related to HPT.And to explore whether the improvement of VD cognitive dysfunction by Yinao Jieyu formula is related to the regulation of thyroid hormone changes.Methods The VD model of rats was selected after the cerebral vascular lesions were caused by the modified version of thread embolization method.After 4 weeks without any intervention,the VD model was screened out by the platform test.The experiment was divided into 4 groups,including normal group,sham operation group,VD model group and VD Chinese medicine group,with 15 rats in each group.VD Chinese medicine group rats was given a gavage of Yinao Jieyu formula,and rats in normal group,sham operation group and VD model group were given a gavage of 0.9%normal saline,continued for 4 weeks.After the final platform test and Morris water maze test were performed by intragastric administration at 1 week and 4 week,6 rats in each group were randomly selected,anesthetised and sacrificed,and HE staining of hypothalamus was performed to observe their pathological morphology.After the last behavioral test at 4 week,8 rats were randomly selected for blood collection from abdominal aorta.The contents of thyroid stimulating hormone(TSH),free triiodothyronine(FT3)and free thyroxine(FT4)in serum were detected by radioimmunoassay.Results The platform test data showed that compared with the normal group,the escape latency time of rats intragastric administration for 1 week and 4 week in VD model group and VD Chinese medicine group was significantly decreased,and the errors number of rats intragastric administration for 1 week was significantly increased(P<0.01).Compared with VD model group,the escape latency time of rats intragastric administration for 1 week and 4 week in VD Chinese medicine group were significantly increased,and t
