摘 要:目的探讨痛泻要方对IBS模型大鼠SP和CGRP表达的影响。方法建立肠易激综合征(irritable bowel syndrome,IBS)模型,将大鼠随机分为正常组、模型组、得舒特组、痛泻要方组。检测结肠转运功能、小肠推进率;大鼠焦虑情况应用高架十字迷宫模型测定;ELISA法检测血浆SP和CGRP的表达;应用Western blot法检测结肠组织SP和CGRP蛋白的表达量。结果与正常组比较,模型组、得舒特组、痛泻要方组大鼠的玻璃小球排出时间明显缩短(P<0.01);与模型组比较,得舒特组、痛泻要方组的排出时间明显延长(P<0.01);得舒特组和痛泻要方组比较差异无统计学意义(P>0.05)。与正常组比较,模型组、得舒特组、痛泻要方组大鼠的小肠推进率明显升高(P<0.01);与模型组比较,得舒特组、痛泻要方组的小肠推进率明显降低(P<0.01);得舒特组和痛泻要方组比较差异无统计学意义(P>0.05)。模型组大鼠OE%、OT%相对于正常组显著降低,差异有统计学意义(P<0.05),在得舒特组和痛泻要方组又得以恢复,舒特组和痛泻要方组与模型组比较,差异有统计学意义(P<0.05)。与正常组比较,SP和CGRP的表达在模型组显著上升(P<0.05)。与模型组比较,得舒特组和痛泻要方组SP和CGRP的表达水平显著降低(P<0.05)。结论痛泻要方在治疗IBS中,有效地改变了肠道转运功能,通过调节SP和CGRP信号通路的表达平衡了炎症因子的释放,通过控制焦虑情绪改变了SP和CGRP的表达,进一步减轻炎症反应。Objective To investigate the effect of Tongxie Decoction on SP and CGRP expression in irritable bowel syndrome(IBS)rats.Methods The IBS models were established,and the rats were randomly divided into normal group,model group,deshute group,and Tongxie Decoction group.Colon transport function and the rate of small intestine propulsion were determined;The anxiety in rats were determined by elevated cross maze model;The expression of plasma SP and CGRP were determined by ELISA method;and the expression of colon tissue SP and CGRP protein were determined by Western blot.Results The excretion time of vitreous pellets was significantly shorter in the model group,Deshute group and Tongxie Decoction group,compared with the normal group(P<0.01);and significantly longer in the Deshute group and Tongxie Decoction group,compared with the model group(P<0.01).While there was no statistical significance in the Deshute group and Tongxie Decoction group(P>0.05).Compared with the normal group,the small intestine propulsive rate of rats in the model group,Deshute group and Tongxie Decoction group increased significantly(P<0.01).And significantly decreased in the Deshute group and Tongxie Decoction group,compared with the model group(P<0.01).While there was no statistical significance in the Deshute group and Tong-xie Decoction group(P>0.05).The OE%,OT%of rats in the model group were significantly lower than that in the normal group(P<0.05),but recovered again in the deshute group and the tongxie Decoction group.Compared with the model group,there were significant increase in the Deshute group and Tongxie Decoction group(P<0.05).The expression of SP and CGRP in plasma increased significantly in the model group,compared with the normal group(P<0.05).However,there was a significant decrease in the Deshute and Tongxie Decoction groups compared with the model group(P<0.05).The expression of SP and CGRP proteins increased significantly in the model group,compared with the normal group(P<0.05).However,the expression levels of SP and CGRP p
