鸡西产黄芩药材及其须根的质量评价与溯源研究

摘  要:目的:建立黄芩药材和须根的质量控制方法,考察生长年限和施肥情况对鸡西产黄芩药材和须根中主要成分含量的影响。方法:采用HPLC,C 18色谱柱;以甲醇-0.4%磷酸水溶液为流动相进行梯度洗脱;检测波长277nm;柱温25℃;流速1.0mL·min^(-1)。结果:黄芩苷、黄芩素和汉黄芩素的线性范围分别为2.705~108.2μg·mL^(-1)、0.5465~21.86μg·mL^(-1)和0.4660~18.64μg·mL^(-1),r≧0.9996;平均回收率分别为98.78%、97.17%和97.16%,3年生且施加有机肥黄芩药材及须根各项指标含量较高。结论:本法操作简便,专属性强,准确度高,可用于黄芩药材与须根的质量评价,鸡西地区栽培黄芩种植年限选择3年并施加有机肥有利于提高黄芩的品质且能节约种植成本。Objective:To establish a quality control method for Scutellariae Radix and its fibrous roots,and to investigate the effects of growth years and fertilization on the contents of main components in them from Jixi.Methods:The contents were determined by HPLC:C18 column;gradient elution with methanol -0.4% phosphoric acid as mobile phase;detection wavelength was 277nm;column temperature was 25℃;flow rate was 1.0mL·min^(-1).Results:the linear ranges of baicalin,baicalein and wogonin were 2.705~108.2μg·mL^(-1),0.546~21.86μg·mL^(-1) and 0.4660~18.64μg·mL^(-1) respectively,r≧0.9996.The average recoveries were 98.78%,97.17% and 97.16% respectively.The contents of 3-year-old organic fertilizer sample were higher than in other samples.Conclusion:The method is simple,specific and accurate.It can be used for the quality evaluation of Scutellariae Radix and fibrous roots.The Triennial Scutellariae Radix in Jixi should be selected and organic fertilizer should be applied to improve the quality and save the planting cost.
