摘 要:目的探讨热暴露下大鼠胸主动脉结构变化及胸主动脉内皮细胞钟基因BMAL1和细胞周期蛋白表达水平的改变。方法选用雄性SD大鼠20只,随机分为正常对照组和热暴露组各10只。HE染色观察大鼠胸主动脉组织病理变化,IHC检测大鼠胸主动脉BMAL1表达情况。体外实验采用大鼠胸主动脉内皮细胞株(RTAEC),分为对照组和热暴露组,体内和体外实验均采用Western blot检测BMAL1及细胞周期蛋白CDK1、CDK4、CDK6、CyclinB1的表达。结果正常组大鼠与热暴露组相比体质量差异无统计学意义(P>0.05);正常对照组大鼠胸主动脉血管管壁完整,热暴露组血管内皮细胞明显缺失,弹性纤维紊乱、断裂;与正常组比较,热暴露组大鼠胸主动脉血管组织BMAL1阳性表达增多,BMAL1、CyclinB1、CDK1蛋白表达均升高(P均<0.05)。体外实验结果显示:与正常组比较,热暴露组大鼠胸主动脉内皮细胞BMAL1、CyclinB1、CDK1蛋白表达均升高(P均<0.05)。结论热暴露可以引起大鼠胸主动脉内皮结构改变;大鼠胸主动脉内皮细胞钟基因BMAL1和细胞周期蛋白CyclinB1、CDK1表达上调,提示热暴露可以改变钟基因与细胞周期蛋白的表达水平,此变化可能是通过影响G2/M期蛋白表达来实现的。Objective To investigate the structural changes of thoracic aorta and the expressionlevelsof clock gene BMAL1 and cyclin proteins of thoracic aortic endothelial cells under heat exposure.Methods In vivo,twenty male SD rats were randomly divided into normal control group and heat exposure group,and histopathological changes of rat thoracic aorta were observed by HEstaining,BMAL1 expression of rat thoracic aorta was detected by IHC.In vitro,rat thoracic aortic endothelial cells(RTAECs)were also divided into normal control group and heat exposure group.In vivo and in vitro,Western blot was used to detect the expression levels of BMAL1,CDK1,CDK4,CDK6 and CyclinB1.Results There was no significant difference in body weight between normal control group and heat exposed group(P>0.05).The vascular walls of thoracic aorta in normal control group were intact,while the vascular endothelium in heat exposure group were obviously lost,the elastic fibers were disordered and ruptured.Compared with the normal control group,the positive expression of BMAL1in thoracic aorta of rats exposed to heat increased,and proteins of BMALI,CyclinB1 and CDK1 showed the same trends(P all<0.05).Compared to normal group,Western blot showed that expression levels of BMALI,CyclinB1 and CDK1 protein increased in thoracic aotic endothelial cells in heat exposure group(P all<0.05).Conclusion Heat exposure can cause changes in endothelial structure of rat thoracic aorta.The up-regulation of BMAL1,CyclinB1 and CDK1 in rat thoracic aortic endothelial cells suggests that heat exposure can change the expression levels of clock genes and cyclin,which may be achieved by affecting the protein expression of G2/M phase.
